头孢菌素C产生菌顶头孢霉理性化筛选的研究

摘    要:顶头孢霉82-128菌株改良过程中,除常规的随机筛选外,还采用了理性化筛选,以提高头孢菌素C的产量,方法为筛选营养缺陷型回复突变株及抗重金属离子变株.试验结果表明理化筛选优于随机筛选.正突变频率前者为50.87%,后者为38.25%.变株幅度,前者产量高于亲株200%的占6.16%,高于亲株300%的占2.84%;后者产量高于亲株200%的占0.258%,高于亲株300%的未出现.

Abstract:This paper deals with the rational selection in Cepalosporium acremonium. The rational screening was used to improve Cephalosporium acremonium 82-123 to increase its antibiotic productivity. The main processes in the method are screening mutants of auxotrophies reversion and resistance to metal ions. The results showed that the rational selection (A) was better than random selection(B). The positive frequencies of mutation were A, 50.87% and B, 38.25%. The ranges of mutation rate were as follows: In (A), mutants whose productivity 200 and 300% higher than parent strain, (as control), were rated 6.16 and 2.84% respectively. In (B), mutants whose productivity 200 and 300% higher than parent strain, were rated 0.258 and 0% respectively. This results indicated that (A) not only higher in positive frequency of mutation than (B), but also the range of mutation as well.
