摘    要:目的优选在以葡萄糖为对照时,苯酚-硫酸法测定多糖含量的显色方式.方法采用正交试验法.结果对正交试验结果进行直观分析和方差分析,优选显色方式为:取待测溶液2.0mL,加入5%苯酚液1.0mL,混匀,迅速滴加浓硫酸7.5mL,摇匀,室温放置1 5 min,然后置冷水中冷却30 min,在490 nm波长处测定.结论优选的显色方式操作更简便、易行.

Abstract:Objective To optimize the way of coloration in the process of determining polysaccharide content by phenol-sulfuric acid method, using only glucose as the standard. Method By orthogonal test. Results After synthesizing the results of direct analysis and variance analysis, the optimum way of coloration was obtained as follows: 1.0 mL 5% phenol solution was added to 2.0 mL testing solution and mixed. Then 7.5 mL concentrated sulfuric acid was added quickly drop by drop. After set at ambience for 15 min and cooled following in cold water for 30 min, the solution was detected at wavelength of 490 nm. Conclusion The optimum way of coloration can be easily controlled and operated.
