黄连地黄汤对糖尿病性心肌病大鼠心肌CTGF及FN1表达的影响

摘    要:[目的] 研究黄连地黄汤对糖尿病性心肌病的防治作用。[方法] 80只SPF级SD大鼠分为空白对照组、模型组、黄连地黄汤低剂量组(2.7 g/kg生药)、黄连地黄汤中剂量组(5.4 g/kg生药)、黄连地黄汤高剂量组(10.8 g/kg生药)、二甲双胍(90 mg/kg)合并苯那普利(0.9 mg/kg)对照组。除空白对照组外其余大鼠先用高脂高糖饲料喂养6周,然后各组造模大鼠腹腔注射浓度为44 mg/kg的链脲佐菌素(STZ)。3 d后检测空腹血糖,选择血糖大于11.1 mmol/L的2型糖尿病模型。血糖符合标准的大鼠继续边喂饲高脂饲料边给药12周。末次给药1 h后测空腹血糖,然后各组大鼠用5%水合氯醛(6 mL/kg)麻醉,腹主动脉取血,检测血清低密度脂蛋白胆固醇(LDL-C)、总胆固醇(TC)、超氧化物歧化酶(SOD)、丙二醛(MDA),取心脏用福尔马林溶液固定,用于苏木精-伊红(HE)染色及免疫组化检测心肌细胞结缔组织生长因子(CTGF)和纤维连接蛋白抗体(FN1)表达。[结果] 研究结果表明黄连地黄汤可降低糖尿病性心肌病模型大鼠空腹血糖值、LDL-C、TC含量及心肌细胞结缔组织生长因子的表达,增加其抗氧化能力及纤维连接蛋白的表达。[结论] 黄连地黄汤对糖尿病性心肌病模型大鼠心肌细胞具有保护作用。

Effect of Huanglian Dihuang Decoction on the expression of CTGF and FN1 in myocardium of rat with diabetic cardiomyopathy

Abstract:[Objective] To study the preventive and therapeutic effects of Huanglian Dihuang Decoction on diabetic cardiomyopathy.[Methods] Eighty SPF SD rats were divided into blank control group,model control group,Huanglian Dihuang Decoction low-dose group (2.7 g/kg crude drug),Huanglian Dihuang Decoction medium-dose group (5.4 g/kg crude drug),and Huanglian Dihuang Decoction high-dose group (10.8 g/kg crude drug),metformin combined with benazepril control group (90 mg/kg,0.9 mg/kg). Except the blank control group,the other rats were first fed with high-fat and high-sugar diet for 6 weeks,and then the rats in each group were injected with streptozotocin (STZ) at a concentration of 44 mg/kg intraperitoneally. After 3 days,fasting blood glucose was measured,and a type 2 diabetes model with a blood glucose greater than 11.1 mmol/L was selected. Rats whose blood glucose met the standard continued to be fed with high-fat diet for 12 weeks. Fasting blood glucose was measured 1 hour after the last administration,and then rats in each group were anesthetized with 5% chloral hydrate (6 mL/kg),blood was taken from the abdominal aorta,and serum LDL-C,TCH,SOD,and MDA were measured. Hearts were fixed with formalin and used for HE staining and immunohistochemical detection of CTGF and FN1 expression in cardiomyocytes.[Results] The results of this study indicate that Huanglian Dihuang Decoction can reduce fasting blood glucose,LDL-C and TC content and the expression of connective tissue growth factor of cardiomyocytes in diabetic cardiomyopathy model rats,and increase its antioxidant capacity and fibronectin expression.[Conclusion] Huanglian Dihuang Decoction has protective effect on cardiomyocytes in diabetic cardiomyopathy model rats.
