COX-2在肝细胞肝癌中的表达及其与细胞凋亡的关系

目的 探讨肝细胞癌(HCC)中环氧化酶-2(COX-2)的表达与细胞凋亡的关系及其可能的机制.方法 选用44例HCC组织制作组织芯片,应用免疫组织化学S-P法检测癌组织COX-2、caspase-3和survivin的表达情况.应用末端标记法(TUNEL)检测细胞凋亡指数(AJ).结果 HCC中COX-2蛋白阳性率为52.3%,其表达与AJ呈显著负相关r=-0.354,P＜0.05),与caspase-3蛋白表达也呈显著负相关(r=-0.447,P＜0.01),但与survivin蛋白的表达呈显著正相关(r=0.394,P＜0.01).AI与caspase-3呈显著正相关(r=0.381,P＜0.05),但与survivin呈显著负相关(r=-0.458,P＜0.01).结论 本研究证实了HCC中COX-2的抗肿瘤细胞凋亡作用;其机制可能是通过上调肿瘤细胞survivin表达,同时下调caspase-3表达途径实现的.

RELATIONSHIP BETWEEN THE EXPRESSION OF COX-2 AND CELL APOPTOSIS IN HEPATOCELLUAR CARCINOMA

Objective To study the relationship between the expression of cyclooxygenase-2(COX-2) and apoptosis in hepatocelluar carcinoma(HCC),and to investigate its possible mechanisms.Methods The protein expression of COX-2,caspase-3 and survivin was detected by immunohistochemical method,and cell apoptosis was examinated by TUNEL technique in tissue microarrys constructed with 44 cases of HCC.Results COX-2 expressed in 52.3% of 44 HCC cases,which was negatively correlated with apoptotic indices(AI)(r=-0.354,P<0.05) and caspase-3 expression(r=-0.447,P<0.01),but positively correlated with survivin(r=0.394,P<0.01).AI was positively correlated with the expression of caspase-3(r=0.381,P<0.05),but negatively with that of survivin(r=-0.458,P<0.01).Conclusion COX-2 might play a significant role in the anti-apoptosis effect of HCC cells via up-regulating survivin expression and down-regulating caspase-3 expression.
