胃腺癌组织CD151、Cav-1与MT1-MMP的表达及其临床意义

摘    要:目的:探讨胃腺癌组织分化抗原151(CD151)、小凹蛋白-1(Cav-1)与膜型基质金属蛋白酶-1(MT1-MMP)的表达变化及其临床意义。方法选取2014年5月至2015年5月我院消化外科胃腺癌手术患者65例,术中采集胃腺癌组织标本65例作为研究组,癌旁正常胃黏膜组织标本65例作为对照组,采用SP免疫组化方法检测CD151、Cav-1与MT1-MMP蛋白表达水平。结果研究组CD151与MT1-MMP蛋白阳性表达率分别为78.46%和80.00%,明显高于对照组的6.15%和12.31%,Cav-1蛋白阳性表达率为52.31%,明显低于对照组的80.00%,两组比较差异均具有统计学意义(P<0.05);CD151、Cav-1与MT1-MMP蛋白表达与组织学分化程度、临床分期和淋巴结转移等临床病理特征具有明显的相关性,其中低分化及其未分化胃腺癌、中晚期胃腺癌及其有淋巴结转移胃腺癌CD151与MT1-MMP蛋白阳性表达率显著增高,Cav-1蛋白阳性表达率显著降低,不同临床病理特征胃腺癌组织CD151、Cav-1与MT1-MMP蛋白阳性表达率比较差异均具有统计学意义(P<0.05);经Pearson相关性分析显示,CD151、MT1-MMP与Cav-1表达之间具有明显的负相关(r=-0.521,-0.501,P<0.05),CD151与MT1-MMP表达之间具有明显的正相关(r=0.547,P<0.05)。结论胃腺癌组织CD151与MT1-MMP表达明显上调,Cav-1表达明显下调,三者均与临床病理特征具有紧密的关系。

Abstract:Objective To investigate the expression of CD151, caveolin-1 (Cav-1) and membrane type matrix metalloproteinase-1 (MT1-MMP) and its clinical significance. Methods We recruited 65 patients of gastric cancer from May 2014 to May 2015 in our hospital. Gastric cancer specimens (n=65, the study group) were collected intraoperatively as study group, and the adjacent normal gastric mucosa tissues (n=65, the control group) were collected as control group, Expression of CD151, Cav-1 and MT1-MMP protein were detected by immunohistochemical method. Results The pos-itive expression rates of CD151 and MT1-MMP in the study group were significantly higher than those in the control group (78.46%vs 6.15%, 80.00%vs 12.31%), while the positive expression rate of Cav-1 protein was significantly lower than that in the control group (52.31%vs 80.00%), all with statistically significant differences (P<0.05). CD151, Cav-1 and MT1-MMP protein expression had significant relevance with clinicopathological features of histological differentia-tion, clinical staging and lymph node metastasis. In patients with poorly and undifferentiated gastric adenocarcinoma, ad-vanced gastric adenocarcinoma, and gastric adenocarcinoma complicating lymph node metastasis, the positive expres-sion rate of CD151 and MT1-MMP protein was significantly increased, and the Cav-1 protein positive expression rate was significantly decreased. The positive expression rates of CD151, Cav-1 and MT1-MMP protein showed statistically significant difference among patient groups with different clinicopathological characteristics (P<0.05). Pearson correla-tion analysis showed that, there were significant negative correlations between CD151, MT1-MMP and Cav-1 (r=-0.521,-0.501, P<0.05), and a significant positive correlation between CD151 and MT1-MMP (r=0.547, P<0.05). Conclusion In gastric cancer tissues, CD151 and MT1-MMP expression are up-regulated and Cav-1 expression is sig-nificantly down-regulated, which has close relationship with clinicopathological characteristics.
