急性脑梗塞患者脑脊液的高水平髓鞘素抗体分泌细胞

摘    要:为确定急性脑梗塞是否在对髓鞘素碱性蛋白,髓鞘素结合糖蛋白和含脂质蛋白的B细胞免疫应答,采用酶联免疫斑点技术检测了发病后4周内的ACI和其它神经疾病的对照组患者外周血和脑脊液的MBP,MAG和PLP抗体分泌细胞,发现ACI和OND外周血检出的IgG,IgA,IgM三种表型MBP,MAG和PLP抗体分泌细胞数无显著差异,但ACI患者CSF的IgF型MBP,MAG和PLP抗体分泌细胞数呈显著性增高。

Abstract:To demonstrate whether B cell responses to myelin antigens in acute cerebral infarct (ACI),the numbers of cells in peripheral blood and cerebrospinal fluid (CSF) secreting antibodies to myelin basic protein (MBP),myelin associated glucoprotein (MAG) and myelin proteolipid protein (PLP) were examined by using enzyme lindek immunospot (ELISPOT) assays in ACI patientss (whithin 4 weeks after onset) and for comparison,in patients with other noninflammatory neurological diseases (OND).Cells secreting antibodies to MBP,MAG and PLP of IgG,IgA and IgM isotypes were detected in peripheral blood from the patients with ACI and OND,and there was no difference between the 2 patients' groups.In contrast,CSF from patients with ACI contatined elevated numbers of cells secreting IgG antbodies to MBP,MAG and PLP.The findings suggest that the brain damage associated with acute ischemia lead to in vivo strong B cell activation and responses to myelin antigens within the central nervous system.The role of such B cell autoreactivities in ACI remains to be settled.
