甲氨蝶呤对寻常型银屑病患者外周血MMP-2表达的调控作用

目的：探讨甲氨蝶呤（methotrexate，MTX）对寻常型银屑病患者外周血中基质金属蛋白酶2（matrix metalloproteinase 2，MMP-2）表达的调控作用。方法：采用酶联免疫吸附试验（ELISA）技术检测19例寻常型银屑病外周血治疗前，MTX治疗8周后及19例正常对照者外周血MMP-2的表达。结果：MMP-2在寻常型银屑病外周血中治疗前的表达水平为（268．94±43．15）ng／mL，经过MTX治疗8周后的表达水平为（228．67±50．32）ng／mL，正常对照者的表达水平为（213．27±53．04）ng／mL。MMP-2在寻常型银屑病外周血治疗前的表达水平明显高于治疗后及正常对照者的表达水平（P〈0．01），治疗后的表达水平与正常对照者的表达水平差异无统计学意义（P〉0．05）。结论：MTX能抑制银屑病患者外周血MMP-2的表达，这可能是MTX治疗银屑病的作用机制之一。

Studyof Methotrexate on Matrix Metalloproteinase 2 Expression in Patient with Psoriasis Vulgaris

Objective：To investigate methotrexate on matrix metalloproteinase 2 （MMP -2） expression in patients with psoriasis vulgaris. Methods：Nineteen adult patients with psoriasis vulgaris and 19 health control were sequentially enrolled in this study. Serum level of MMP -2 were measured by enzyme linked immunosorbent assay （ELISA） in study group before and after treatment with MTX for 8 weeks and. Results：The serum level of MMP -2 before andafter treatment in study group and health control were 258.94 ±43.15 ng/mL, 228.57 ±50.32 ng/mL and 213.27 ± 53.04 ng/mL, respectively. The serum levels of MMP - 2 before treatment was significantly higher than that after treatment and health control（P 〈0.01 ）. The serum levels of MMP-2 were no statistics significant between treatment group and health control group （P 〈 0.01 ）. Conclusion： Theses results were indicated that MTX might inhabit the over expression of MMP -2 in patients with psoriasis vulgaris. These changes could be part of the pathogenesis of MTX on psoriasis.
