短尾蝮蛇毒磷脂结合抗凝蛋白对动物血栓的抗栓作用

摘    要:目的从短尾蝮蛇毒中分离纯化磷脂结合抗凝蛋白(PBAP),研究其对动物血栓的抗栓作用。方法用Chandler法制备家兔体外血栓和半体内血栓模型;用Reyers法制备大鼠静脉血栓模型;用Nowork法制备家兔肺动脉栓塞模型。测定PBAP对家兔体外血栓、半体内血栓、肺动脉栓塞和大白鼠静脉血栓的抗栓效应。结果短尾蝮蛇毒PBAP对家兔体外血栓、半体内血栓、肺动脉栓塞和大白鼠静脉血栓干湿重均有明显减轻作用,与生理盐水对照组比较,差异均有统计学意义(P<0.05或P<0.01)。结论PBAP抗凝蛋白对动物血栓具有明显的抗栓作用,具有潜在的临床应用价值。

Abstract:Purpose To study thrombolytic effect of phospholipid-binding anticoagulation protein(PBAP)purified from Agkistrodon halys brevicaudus venom on animal thrombosis.MethodsBy means of pulmonary embolism model in rabbits,venous thrombosis model in rats and Chandler′s mothod in vitro.The phospholipid-binding anticoagulation protein(PBAP) was given by intravenous injection at 0.2,0.4,0.8 mg/kg three different doses respectively.Normal saline was used as negative control and thrombolytic effects was observed.ResultsIt was showed that moist weight of venous thrombosis and pulmonary embolism were markedly lightened and has statistical significance,comparative with normal saline.ConclusionThe phospholipid-binding anticoagulation protein(PBAP) from Agkistrodon halys brevicaudus venom has obvious thrombolytic effect on experimental thrombosis of animals.It was effective modern antithrombotic pharmaceutics with potential clinical applied values.
