金黄色葡萄球菌感染乳腺炎小鼠T淋巴细胞亚群水平变化

摘    要:目的探讨金黄色葡萄球菌感染乳腺炎小鼠T淋巴细胞亚群水平变化。方法采用金黄色葡萄球菌乳头管灌注法制备乳腺炎模型小鼠;将小鼠分为空白组(B)、对照组(C)、感染24 h(M1)、感染72 h(M2)、感染120 h(M3)5个试验组,于试验结束时观察各试验组小鼠健康状况、乳腺组织病理变化,并测定各试验组小鼠体温、体质量、外周血细胞分类数量、乳腺组织菌落数,测定白细胞介素-6(IL-6)、IL-17A、转化生长因子-β1(TGF-β1)、肿瘤坏死因子-α(TNF-α)蛋白表达水平。结果模型小鼠(M1、M2、M3)的健康状况、病理切片观察和乳腺组织学指标均低于B、C组,体质量、细菌菌落数、炎症细胞数目与B、C组比较,差异有统计学意义(P<0.05),且随着感染天数增多,细菌菌落数与炎症细胞数目呈增长趋势(P<0.05);模型小鼠(M1、M2、M3)乳腺组织IL-6、IL-17A、TGF-β1、TNF-α蛋白表达水平和Th17/Treg细胞亚群数目及比例高于B、C组,差异有统计学意义(P<0.05)。结论金黄色葡萄球菌感染致产后小鼠实验性乳腺炎模型小鼠的乳腺状况、相关细胞因子表达水平、Th17/Treg细胞亚群数目及比例随感染天数增加。

Abstract:OBJECTIVE To explore the changes of T lymphocytes subsets induced by Staphylococcus aureus bacterial infection in mice with mastitis.METHODS The mastitis mice models were prepared by perfusion of S.aureus nipple tube;the mice were divided into 5 test groups:the blank group(B),the control group(C),infected for 24 h(M1),infected for 72 h(M2)and infected for 120 h(M3).At the end of the test,the health status and pathological changes of the breast tissues of the mice were observed,the body temperature,body weight,classification number of peripheral blood cells and bacterial colony counts in the breast tissues were determined,and the levels of interleukin-6(IL-6),interleukin-17 A(IL-17 A),transforming growth factor-β1(TGF-β1)and tumor necrosis factor-α(TNF-α)were detected.RESULTS The health status,pathological indexes and mammary histological indexes of the model groups(M1,M2,and M3)were lower than those of the group B and the group C.There were significant differences in the body weight,bacterial colony counts and number of inflammatory cells between the model groups and the group B and the group C(P<0.05);the bacterial colony counts and number of inflammatory cells showed upward trends with the increase of number of infection days(P<0.05).The levels of IL-6,IL-17 A,TGF-β1 and TNF-αin breast tissues and percentages of Th17/Treg cells subsets were significantly higher in the model groups(M1,M2,M3)than in the group B and the group C(P<0.05).CONCLUSION The status of mammary glands,levels of related cytokines and percentages of Th17/Treg subsets of the experimental mastitis mice induced by S.aureus infection are increased with the number of infection days.
