| 摘 要： | 目的研究灵芝三萜的最佳提取方法,并测定其总三萜的含量。方法采用正交实验方法,用硅胶柱进行总三萜分离,通过紫外-可见分光光度法测定总三萜含量。结果最佳提取方法为用5倍量95%乙醇回流提取3次,每次1.5 h;用30倍量硅胶装柱,以石油醚-乙酸乙酯(9:1和7:3)进行洗脱;经分离得到针状晶体Ⅰ和Ⅱ,测定该品种中灵芝总三萜含量为9.07%。结论本提取方法稳定可行,可作为灵芝三萜工业化生产的参考。

| Abstract: | Objective To design the best out of Ganoderma triterpenoids extract programs and separation methods,and measured the total content of triterpenoid.Methods Filter through the orthogonal extraction method,using silica gel column for the total separation of triterpenoids by ultraviolet-visible spectrophotometry triterpenoid content.Results The optimum extraction method is to use f ive times the amount of 95% ethanol was extracted three times for 1.5 hours each time;loaded with 30 times the amount of silica gel column with petroleum ether-ethyl acetate（9：1 and 7：3） for elution;by ultraviolet-visible spectrophotometry in order to ursolic acid as a reference substance,the total triterpene content of Ganoderma lucidum were determined.Conclusion The extraction method is stable and feasible,and Ganoderma triterpene content was to 9.07%. |
