细菌感染大鼠下丘脑及垂体ACTH样和β-内啡肽样免疫阳性细胞的相关变化

摘    要:取正常Wistar大鼠,雌雄兼用.老龄鼠(20个月)9只,壮龄鼠(8个月)10只.通过腹膜腔注入致病量福氏痢疾杆菌F1b(活菌492万个/克体重),24h后,处死取材;常规脱水,石蜡切片;ABC法染色,DAB显色.光镜下观察下丘脑及垂体促肾上腺皮质激素样免疫反应(ACTH-ir)和β-内啡肽样免疫反应(β-END-ir)阳性细胞的形态、分布和计数.图像分析仪测其灰度值.结果:多数鼠第三脑室室周层出现许多β-END-ir强阳性细胞.ACTH-ir阳性细胞数量,感染老龄鼠显著低于感染壮龄鼠,β-END-r阳性细胞数量,感染老、壮龄鼠间无差异.感染老龄鼠弓状核和垂体前叶ACTH-ir阳性细胞灰度显著高于壮龄鼠,垂体前叶β-END-ir阳性细胞灰度则显著低于壮龄鼠.与正常组比较,感染老龄鼠弓状核和垂体前叶的ACTH-ir阳细胞灰度显著上升,壮龄鼠则下降;老龄鼠弓状核β-END-ir阳性细胞灰度上升,垂体则下降,壮龄鼠全上升.提示:ACTH和β-END与抗感染密切相关.

Abstract:In this study, the female and male Wistar rats, 9 aged (twenty-month-old) and 10 adult (eight-month-old) were used. Infected .with Shengilla Flexner (Fib) injected into peritoneal cavity, the rats were survived for 24 hours and were sacrific d. The blocks containing hypothalamus and the pituitary were obtained, dehydrated and embedded in Paraffin(50~52℃ ). The sections(7 μm) were stained with ABC technique and coloured by DAB. The morphology, distribution and counting of ACTH-ir and β-END-ir cells in the rat hypothalamus and the pituitary were obaerved and measured under microscope. The light values of positive cells were measured with Image Analyser. We had found that many β-END-ir deep colour positive neurons appeared in the periventricular layer of the Ⅲ ventricle in most of the rats, and there were not in the normal rats. It is suggested that these may be potential β-END-ir neurons. The numbers of ACTH-ir neurons in the aged rats are smaller than those in the aged and adult rats, but the counting of β-END-ir neurons, both in aged and adult rats, are similar. The light values of ACTH-ir and β-END-ir neurons of the arcuate nucleus in the aged rats were higher than those in the adult rate. The light values of ACTH-ir cells of anterior lobe in the aged rat pituitary was higher than that in the adult rat, while the light value of β-END-ir cells in the aged rat lower than that is in the adult rat. As compared with the normal group, the light value Of ACTH-ir neurons of arcuate nucleus in the infected aged rats was obviously incresed, on the other hand, it was decreaaed in the infected adult rats. In terms of the light values rats of β-END-ir neurons of arcuate neucleus, both the aged and adult rats in the infected group were obviously higher than those in the normal group. The light value of ACTH-ir cells of pituitary anterior lobe in the infected aged rats was increased, which was decreased in the infected adult rats. The light value of β-END-ir cells of pituitary anterior lobe in the infected aged rats was decreased, white it was increased in the infected adult rats. The results suggested that both ACTH and β-END are correlated with infection and immune process.
