戊四氮点燃过程中大鼠海马 BMP4表达与神经增殖关系

摘    要:目的探讨戊四氮(PTZ)点燃过程中大鼠海马骨形成蛋白4(bone morphogenetic protein-4,BMP4)的表达变化与神经增殖的关系。方法将成年大鼠分为对照组与模型组,模型组大鼠又根据在点燃中的不同时相点分为9组。用免疫组织化学与原位杂交的方法检测海马齿状回BMP4mRNA与BrdU阳性细胞数的变化。结果正常成年大鼠BMP4阳性细胞主要分布于齿状回的门区、颗粒下层、CA3、CA1区。BrdU阳性细胞主要分布在齿状回颗粒下层。BMP4阳性细胞与BrdU阳性细胞在点燃过程中均明显增加,呈明显正相关,点燃后2月降至基线水平。结论BMP4可能通过影响成年大鼠海马神经发生在PTZ点燃过程中起重要作用。

Abstract:Objective To investigate the relationship between the expression of bone morphogenetic protein-4 (BMP4) in the hippocampus and the neurogenesis during the pentylenetrazol(PTZ) kindling process of the adult rats.Methods Rats were divided into two groups:the control group and the PTZ kindling group.And the PTZ kindling group includes 9 subgroup according to different time phase.The changes of the BrdU labelled cells and BMP4 mRNA-positive cells of the hippocampus and denate gyrus was observed by immunohistochemical and in situ hybridization methods.Results The BMP4 mRNA-positive cells were mainly distributed in the hilus and subgranular zone of dentate gyrus and CA_3 and CA_1 of the hippocampus of adult rats.The BrdU labelled cells were mainly distributed in the subgranular zone.The numbers of BMP4 mRNA-positive cells and BrdU labelled cells increased significantly during PTZ kindling process and decreased to base line 2 months later.There were positive correlation between the number of BMP4 mRNA-positive cells and the number of BrdU labelled cells.Conclusion BMP4 likely plays an important role in the onset of seizures through adult neurogenesis of the hippocampus induced by pentylenetrazol.
