摘    要:目的:观察大豆皂甙对人肝癌QGY-7703细胞的生长抑制作用。方法:大豆皂甙作用于肝癌QGY-7703细胞,用MTT法、荧光倒置显微镜观察等研究细胞生长与细胞凋亡,激光共聚焦扫描显微镜(CLSM)观察细胞凋亡及细胞内Ca2+浓度变化。结果:MTT实验表明0.1~0.8mg/ml大豆皂甙对QGY-7703细胞增殖有明显抑制作用。荧光显微镜和CLSM观察到细胞核浓缩等典型的细胞凋亡形态特征。CLSM检测还表明大豆皂甙使肝癌细胞胞内Ca2+浓度显著增大。结论:大豆皂甙对人肝癌QGY-7703细胞有生长抑制作用,诱导细胞凋亡是这种作用的主要形式,胞内Ca2+浓度升高可能是细胞凋亡的作用机制之一。

Abstract:Objective: To study the growth inhibition and the induction of apoptosis brought about by SS-Ⅱ in human hepatoma cell line QGY-7703. Methods: The anti-proliferative activity of SS-Ⅱ was tested in MTT. Apoptotic cancer cells were determined by the confocal laser scan microscopy(CLSM) and the morphological observation under fluorescence. The mechanism of apoptosis was studied with detection of intracellular Ca2+. Results: MTT showed that SS-Ⅱ could inhibit the growth of QGY-7703 effectively in dose-response and time-response manner. The morphological observation by AO/EB double fluorescent dye staining suggested that SS-Ⅱ could increase the apoptosis of hepatoma cells markedly (P<0.01). CLSM revealed for the first time that SS-Ⅱ induced shrinkage in hepatoma cells and chromatin condensation, suggesting apoptosis. It was observed by CLSM that intracellular free Ca2+ level in QGY-7703 was increased remarkably after treatment of SS-Ⅱ as compared with the controls. Conclusion: Apoptosis of hepatoma cells induced by SS is the important antitumor mechanism of SS and the increment of intracellular free Ca2+ level might participate in the process.
