高效液相色谱法同时测定人血清中卡马西平及地尔硫浓度

目的建立反相高效液相色谱（HPLC）法,以安定为内标,同时测定血清中卡马西平（CBZ）和地尔硫（DZ）的浓度。方法采用Waters产μBondapak C18柱,甲醇-磷酸盐缓冲液（内含0.04%二乙胺,pH 6.82）=70∶30为流动相,紫外检测波长为237 nm。结果CBZ两种浓度（5、10 mg/L）的平均回收率及其日内精密度RSD分别为101.6%、100.1%及0.8%、1.0%。DZ两种浓度（100、200μg/mL）的平均回收率及其日内精密度RSD分别为99.6%、101.1%及2.7%、3.0%。结论HPLC法较灵敏、准确、简便。可用于两药合用时血药浓度及药动学变化的研究。

Simultaneous Measurement of Serum Carbamazepine and Diltiazem by HPLC

Objective To establish reversed phase HPLC method for simultaneous determination of the concentration of carbamazepine（CBZ） and diltiazem（DZ） in serum, using diazepam as internal standard. Methods μBondapak C18 column was used as the stationary phase and methanol-phosphate buffer containing 0.04% diethylamine（70 ： 30,pH 6. 82） and the detective wavelength was 237 nm. Results Within-day precision（RSD）and recoveries of two different CBZ concentration（5,10 mg/L） were 101.6 %, 10.0. 1 % and 0.8 %, 1.0 %, respectively. The RSD and recoveries of DZ（100,200μg/L）were 99.6%, 101.1% and 2.7 % ,3.0% ,respectively. Conclusion The method is sensitive, accurate and simple. It can be used in study of the concertration and the pharmacokinetics of CBZ and DZ used.
