摘    要:目的: 优选蛇黄凝胶的提取工艺。方法: 采用L9(34)正交试验设计,以三羽新月蕨苷A、大黄素和干膏得率为评价指标,在单因素试验基础上,通过综合评分优选蛇黄凝胶的提取工艺。采用HPLC测定三羽新月蕨苷A、大黄素含量,流动相乙腈(A)-0.1%磷酸水(B)-水(C)梯度洗脱(0~40 min,10%~70% A,10% B,80%~20%C),检测波长226 nm。结果: 最佳提取工艺条件为加14倍量70%乙醇提取3次,每次30 min;大黄素、三羽新月蕨苷A平均提取量依次为0.37,0.76 mg·g-1结论: 优选的提取工艺稳定可行,对蛇黄凝胶的生产工艺具有一定的指导和参考意义。

Abstract:Objective: To optimize extraction technology of Shehuang gels by orthogonal test. Method: L9(34) Orthogonal test was performed with extraction amounts of triphyllin A and emodin,dry extract yield as indexes,based on single factor tests,extraction process of Shehuang gels was optimized by multi-index comprehensive evaluation method.Contents of triphyllin A and emodin were determined by HPLC,mobile phase of acetonitrile(A)-0.1% phosphoric acid(B)-water(C) with a gradient elution program(0-40 min,10%-70% A,10% B,80%-20%C),detection wavelength 226 nm. Result: Optimal extraction conditions were as follows:extracted 3 times with 14 times the amount of 70% ethanol,30 min for each time;Average extraction amounts of triphyllin A and emodin were 0.37,0.76 mg·g-1. Conclusion: This optimized technology was stable and feasible,which had some guidance and reference value for production of Shehuang gels.
