共培养的树突状细胞与CIK细胞治疗结肠癌血源性肺转移的实验研究

目的 观察树突状细胞(dendritic cells DCs)与同源CIK(cytokine induced killer)细胞共培养后治疗转移性肺癌的可能性。方法 将Lovo结肠癌细胞经尾静脉注射给Balb／c裸鼠建立转移性肺癌模型，应用树突状细胞与同源CIK细胞进行免疫治疗。结果 经尾静脉注射的Lovo结肠癌细胞可播散致肺，导致荷瘤裸鼠死亡。应用树突状细胞和CIK细胞进行免疫治疗，可以抑制转移病灶的形成及延长荷瘤裸鼠的生存期。结论 树突状细胞可增强CIK细胞抗肿瘤效应，共培养DC—CIK细胞有效抑制血源性转移肿瘤的生长，为临床过继免疫治疗的应用提供理论基础。

The experimental study of co-cultured dendritic cells and CIK cells for treating lung hematogenous metastasis of colon cancer

Objective To determine the feasibility of treatment with co-cultured dendritic cells (DC) and CIK cells for lung hematogenous metastasis of colon cancer. Methods The hematogenous metastasis model was established by venous injection of Lovo colon cancer cells in Balb/c nude mice. Then the nude mice were treated with co-cultured dendritic cells and CIK cells. Results The venous injection of Lovo colon cancer cells resulted in lung metastasis and death in the control mice. In contrast, treatment with CIK cells or co-cultured DC-CIK cells inhibited tumor metastasis and rescued the tumor-bearing mice from death. Conclusion The co-cultured dendritic cells and CIK cells could enhance the anti-tumor activity of CIK cells, which were potent in the control of tumor hematogenous metastasis. The result provides an experimental basis for clinical application of DC-CIK cells as adoptive immunotherapy.
