丹参对体外高糖环境中大鼠系膜细胞活性氧抑制作用的研究

摘    要:目的:观察丹参对高糖环境中系膜细胞所产生的活性氧(ROS)及转化生长因子β1(TGF—β1)的抑制作用。方法:将细胞分为以下6组:对照组(C组,普通MEM培养基,含5.6mmol·L^-1葡萄糖);高糖组(H组,30mmol·L^-1高糖MEM培养基);小剂量丹参干预组(S1组,H组+37.5mg·ml^-1丹参);中剂量丹参干预组(S2组,H组+75mg·ml^-1丹参);大剂量丹参干预组(S3组,H组+150mg·ml^-1丹参);单纯丹参组(S组,C组+150mg·ml^-1丹参)。分别采用激光共聚焦和ELISA法检测ROS及TGF—β1水平。结果:H组ROS水平较C组明显增高,分别采用不同浓度丹参干预后,ROS水平均显著降低,并呈剂量依赖性;与H组相比,不同剂量丹参干预组TGF-β1表达均降低,但仅S3组TGF—β1的下降具有统计学意义。与对照组相比,单纯丹参干预组中ROS和TGF—β1水平无明显变化。结论:丹参可减少高糖环境中系膜细胞ROS及TGF—β1的产生,而这可能是丹参发挥肾脏保护作用机制之一。

Abstract:Objective: To investigate the inhibitory effects of Salvia miltiorrhiza on the expression ot reactave oxygen species ( ROS) and transforming growth factor-β1 (TGF-β1 ) induced by high glucose in rat mesangial cells. Method: The mesangial cells were divided six groups : control group ( C, 5.6 mmol ·L - 1 glucose) ; high glucose group ( H, 30 mmol· L - 1 glucose ) ; S1 group ( S1, group H + 37. 5 mg·ml - 1 Salvia miltiorrhiza ) ; S2 group ( S2, group H + 75 mg·ml - 1 Salvia miltiorrhiza) ; S3 group ( S3, group H + 150 Mg· ml - Salvia mihiorrhiza) ;S group ( S, group C + 150 mg·ml -1 Salvia miltiorrhiza). The levels of ROS and TGF-β1 were measured by eonfocal laser scanning microscopy and ELISA respectively. Result: The levels of intraeellular ROS in group H were higher than that in group C ,which were prevented by Salvia mihiorrhiza. And there were dose-dependent. Comparing with group H, TGF-β1 expressions were decreased in three group treated with Salvia miltiorrhiza. Only the decrease of TGF-β1 in group S3 was significant. Comparing with group C ,the levels of ROS and TGF-β1 in group S were not change. Conclusion: Salvia mihiorrhiza may have protective effects on kidney by suppressing the over-expressions of ROS and TGF-β1 in rat mesangial cells induced by high glucose.
