双黄连制剂中连翘苷含量与退热作用相关性研究

目的：研究双黄连制剂中连翘苷的含量与其退热作用的相关性。方法：HPLC法测定连翘苷含量．色谱柱为Diamonsil C18（200mm×4．6mm，5μm），检测波长278nm，流动相为乙腈-水（24：76），流速1．0ml·min^-1，柱温25℃；另大鼠皮下注射酵母菌致热，测定给药后不同时间点的体温，比较各组间体温差异。结果：连翘苷在0．075～1．875μg范围内线性关系良好（r=0．9995），双黄连口服液平均回收率为99．52％，RSD=0．90％（n=9），双黄连片为99．40％，RSD=0．80％（n=9）。其中双黄连口服液（甲）中连翘苷含量最高。各组对酵母菌诱发的发热模型均有解热作用，其中双黄连口服液（甲）解热效果最好。结论：所用方法精密度、重复性良好，结果准确。双黄连制剂中连翘苷含量与药效呈一定相关性。

Study on Relationship Between the Content of Forsythin in Shuanghuanglian Preparations and Antipyretic Effect

Objective： To study on the relationship between forsythin in shuanghuanglian preparations and antipyretic effect. Method： The HPLC method was used to determine the content of forsythin. The Diamonsil C18 column（200 mm × 4. 6 mm,5 μm） was used, mobile phase was acetonitrile-water （24： 76） , detective wavelength was 278 nm, flow rate was 1.0 ml· min^- 1 and column temperature was 25 ℃. Fever was induced by inject yeast into subcutaneous of rats and then temperature was measured at different time after administration to compare the difference between groups. Result： The linear relationship of forsythin was good within the range of 0. 075- 1. 875 μg （ r = 0.999 5 ）. The average recovery of shuanghuanglian orol solution was 99.52% with RSD of 0.90% （ n = 9 ） and shuanghuanglian tablets was 99.40% with RSD = 0. 80% （n = 9）. Shuanghuanglian oral solution （a） had the highest content of forsythin. ALl groups had the antipyretic effect on the fever model induced by yeast,in which shuanghuanglian oral solution （a） was the best. Conclusion： The method is good in precision and repeatability, and the result is accurate. Forsythin in shuanghuanglian preparations was correlation with pharmacodynamie basically.
