大猪屎豆种子中野百合碱的分析

摘    要:目的:建立大猪屎豆种子中野百合碱HPLC含量测定的方法并对其种子及种子水提取物中的野百合碱进行定性与定量的分析。方法:提取和制备野百合碱,采用色谱柱为Diamonsil C18(200mm×4.6mm,5μm);流动相为乙腈-0.01mol·L^-1磷酸二氢钾-三乙胺(5:95:0.1,用磷酸调节pH=3.0);检测波长为205nm;流速为1.0ml·min^-1;柱温为25℃的色谱条件对种子及种子水提取物中的野百合碱进行含量测定。结果:野百合碱在浓度0.102~1.020mg·ml^-1范围内与峰面积呈良好线性关系(r=0.9996,n=5)。野百合碱平均回收率为(n=9)为98.2%,RSD为2.7%;大猪屎豆中野百合碱的含量为2.6%,种子水提取物中的含量为15.0%。结论:本法快速、灵敏、准确,可作为大猪屎豆种子中野百合碱的含量测定方法,并可为进一步毒理学研究提供参考。

Abstract:Objective : To establish an HPLC method for determination of monocrotaline in the seed of Crotalaria assamica Benth. and its decoction. Method: The HPLC separation was performed on a Diamonsil C l8 (200 mm × 4. 6 mm,5 μm) column with a mixture of acetonitrile-0. 01 tool-L^-1 potassium dihydrogen phosphate-triethylamine (5: 95: 0. 1, adjust pH to 3.0 with phosphoric acid) as the mobile phase, at a flow rate of 1.0 ml. L^-l. The detection wavelength was at 205 nm, and column temperature at 25℃. Result: The linear response of monoerotaline ranges from 0. 102-1. 020 mg. ml^-1 ( r = 0. 999 6, n = 5 ). The average recovery of monocrotaline was 98.2% with RSD 2. 7%. The content of monocrotaline in the seed drug and its decoction was 2. 6% and 15% ,respectively. Conclusion: The method is simple, accurate, and reproducible, which can offer reference for the quality control of Crotalaria assamica Benth. and its toxicological study.
