地知复方治疗糖尿病的实验研究及处方筛选

目的：探讨治疗糖尿病中药复方对早期糖尿病小鼠的作用机制并对各方进行筛选。方法：造模前禁食13h，一次性腹腔注射链脲佐菌素170mg·kg^－1造模成糖尿病小鼠（血糖≥13．6mmol·L^－1），并随机分为糖尿病模型组（DM），DM-1治疗组，DM-2治疗组，DM-3治疗组，DM-4治疗组，DM-5治疗组，DM-6治疗组。2周后，检测各组血清及肾组织一氧化氮（NO）、丙二醛（MDA）含量，计算肾脏器指数。结果：与糖尿病模型组相比，DM-2组肾脏指数降低最为显著，抑制血清NO作用最强；DM-2、DM-5组肾组织NO含量显著降低；DM-2、DM-4、DM-6组血清MDA含量明显降低；DM-2、DM-3、DM-4组肾组织MDA含量降低最显著。结论：糖尿病早期给予地知复方的不同组方有一定的治疗作用，它们具有不同程度的抑制肾脏肥厚增生，抗氧化，抑制高血糖刺激引起的NO升高作用，但是只有方2对上述氧化应激指标的改善均有作用，且作用最强。

Screening and Experiment Study of Dizhi Formulas for Treating Diabetes Mellitus

Objective： To study the mechanisms of different Chinese medicinal formulas for treating early diabetes and choose most effective formulas. Method： Experimental diabetes was established by ip sterptozotocin STZ （ 170 mg·kg^-1 ） in mice after fasted for 13 hours. The mice which blood glucose was over 13.6 mmol·L^-1 were considered and were divided into diabetes model group （DM） ,DM- 1, DM-2,DM-3, DM-4,DM-5 and DM-6 treatment group randomly. After two weeks＇ treatment,the content of NO and MDA in serum, kidney were assayed and the kidney index was calculated. Result： Compared with the model group, the kidney index of DM-2 group was decreased mostly significantly;The content of serum NO was decreased most dramatically in DM-2 group;Formula 2,5 could strongly decreased the content of NO in kidney;Formula 2,4 and 6 decreased the content of M DA in serum remarkably;Formula 2,3,4 could most strongly reduce the content of MDA in kidney. Conclusion： All the Chinese herbal formulas in this study are effective in anti-oxidation,decreasing the content of NO in early stage of DM. But only formula 2 has the strongest effects than the others.
