摘    要:目的:建立玫瑰花中槲皮素、没食子酸的含量测定HPLC方法。方法:Kromasil C18色谱柱(250mm×4.6mm,5μm),槲皮素流动相为甲醇-0.4%磷酸溶液(53:47),检测波长:360nm;没食子酸流动相为乙腈-0.1%磷酸溶液(3:97),检测波长:274nm。结果:槲皮素浓度线性范围12.2~244.4μg·ml^-1,r=0.9999;没食子酸浓度线性范围5.6~112.2μg·ml^-1,r=0.9999。平均回收率为槲皮素99.27%,RSD=2.14%(n=6);没食子酸98.68%,RSD=1.79%(n=6)。结论:方法简便、准确、重复性好,可用于该中药的含量测定。

Abstract:Objective: To establish an HPLC method for determination of the quercetin, gallic acid in Rosae rugosae Flos. Method: The Kromasil C18 (250 mm × 4. 6 mm,5 μm) column was used. The mobile phrase of quercetin consisted of methanol-0. 4% phosphoric ocid solulion (53:47 ) , the detection wavelength was 360nm. The mobile phrase of gallic acid consisted of acetonitrile-0. 1% phosphoric (3:97 ) , the detection wavelength was 274 nm. Result: The linear ranges of quercetin and gallic acid were 12. 2-244. 4μg·ml^-1 (r =0. 999 9) and 5.6-112. 2μg·ml^-1(r =0. 999 9) respectively. The average recoveries were 99. 27% (RSD =2. 14% ,n = 6) of quercetin and 98.68% (RSD 1.79% ,n=6) of gallic acid. Conclusion: The method is simple,accurate and repeatable for the determination of quercetin, gallic acid in Rosae rugosae Flos.
