急性心肌缺血损伤后大鼠穴区NO和NOS的变化及不同频率电针的干预作用

摘    要:目的 观察大鼠急性心肌缺血损伤( AMI)后,经穴一氧化氮(NO)含量和一氧化氮合酶(NOS)活性的特异性变化及不同频率电针内关穴在保护急性心肌缺血损伤时对相关经穴NO含量和NOS活性的特异性影响.方法 30只雄性Wistar大鼠随机分为4组,正常组6只,将18只心肌缺血模型制备成功的大鼠平均分为模型组、低频治疗组和高频治疗组各6只.采用硝酸还原酶法测定NO的含量,比色法测定NOS活性.结果 心肌缺血模型组的内关、天泉、郄门、外关和足三里穴区NO水平和NOS活性较正常大鼠同名穴区均显著降低(P <0.001),高频电针内关治疗后,内关、天泉和郄门穴区NO水平较模型组显著升高(P<0.05),内关、外关和足三里穴区NOS活性较模型组明显升高(P<0.05).结论 高频电针内关可保护心肌缺血损伤的机理可能与其阻抑了内关、郄门和天泉穴区NO含量的降低,提高内关、外关和足三里穴区NOS活性相关.

Abstract:Objective To observe the specific changes of content of nitric oxide(NO) and activity of nitric ocide synthase(NOS) around classic acupoints and the specific influences of electroacupunture in different frequency on NO content and NOS activity around related classic acupoints when electroacupunture was used in Neiguan(PC6) in rats with acute myocardial ischemia(AMI).Methods Male Wistar rats(n=30) were randomly divided into four groups.There were 6 rats in normal group,and other successfully modeled rats(n=18) were divided into model group,group treated with low-frequency electroacupunture(low-frequency group) and group of high-frequency electroacupunture(high-frequency group,each n=6).The content of NO was detected by using nitrate reductase method,and activity of NOS was detected by using colorimetric method.Results In model group,NO content and NOS activity dereased significantly around PC6,Tianquan(PC2),Ximen(PC4),Waiguan(TE5) and Zusanli(ST36) compared with normal group(P<0.001).After the treatment with high-frequency electroacupunture in PC6,NO content increased significantly around PC6,PC2 and PC4 compared with model group(P<0.05),and activity of NOS increased significantly around PC6,TE5 and ST36 compared with model group(P<0.05).Conclusion The protective effect of high-frequency electroacupunture in PC6 may be related to that it can inhibit the decrease of NO content around PC6,PC2 and PC4 and improve the activity of NOS around PC6,TE5 and ST36.
