地塞米松对兔骨髓间充质干细胞存活增殖成骨的影响

摘    要:目的:观察骨髓间充质干细胞(BMSCs)的持续生存能力及地塞米松对其存活、增殖、成骨分化能力的影响。方法:提取兔BMSCs,予不同浓度(0、10-9、10-8、10-7、和10-6mol/L)地塞米松,分别于1、3、5、7 d时观察地塞米松对BMSCs增殖的影响,作用1~4周后检测碱性磷酸酶水平;分别饥饿0~7 d,复苏1、3和7 d后,观察BMSCs的持续生存能力。结果:培养1 d地塞米松10-6mol/L组测得的OD值结果最高,而对照组结果最低,培养3、5、7 d时,地塞米松10-9mol/L组测得的OD值最高;同时,在2~4周10-9mol/L组增加碱性磷酸酶活性最明显;复苏1和3 d,对照组与饥饿1~7 d之间差异有统计学意义(均P<0.05),复苏7 d,各组两两之间差异无统计学意义(均P>0.05)。结论:BMSCs具有较强的持续生存能力,地塞米松对BMSCs有增加存活、促进增殖、成骨分化的作用。

Influence of the dexamethasone on bone marrow mesenchymal stem cells, viability, proliferation and osteogenic differentiation

Abstract:Objective: To study the influence of the dexamethasone on BMSCs,viability,proliferation and osteogenic differentiation.Methods: Rabbit marrow-derived mesenchymal cells are subcultured and according to the different density of DEX,BMSCs were randomly classificated into five groups:control group(no DEX,n=5),and trial groups(DEX density of 10-9、10-8、10-7、10-6 mol/L,n=5).After 1,3,5 and 7 days,The influence of DEX to BMSCs,proliferation was tested with MTT methods.After 1,2,3 and 4 weeks,the osteoinductive symbols of BMSCs were detected,the expression of ALP with quantificational method.And according to different days of the hunger randomly classificated to eight groups: control group(no hunger),and trial groups(be hungered for 0~7 days),then resuscitated for 1 day or 3 days or 7 days.The durable livability of BMSCs was tested with MTT methods.Results: After 1 day,the results of the control group was the lowest,and the results were maximal when the density of DEX is 10-6 mol/L.After 3,5 and 7 days,the results were maximal when the density of DEX is 10-9 mol/L.When the density of DEX is 10-9 mol/L after 2~4 weeks,the results were maximal of ALP.After 1 day and 3 days,resususcitated,there were prominence difference between the control groups and the trial groups which had been hungered for different days,P <0.05.After 7 days,resususcitated,there were no special differences between the control group and the trial groups.Conclusion: There are durable livability of BMSCs,and DEX can increase the viability,accelerate the proliferation and osteogenic differentiation of BMSCs.
