乌龙丹对多梗死性痴呆模型大鼠脑组织20种氨基酸含量的影响

摘    要:背景:乌龙丹改善多梗死性痴呆(multi-infarctdementia,MID)学习记忆功能障碍,其作用机制是否与部分氨基酸含量降低有关?目的:探讨乌龙丹对多梗死性痴呆模型大鼠学习记忆功能和脑组织20种氨基酸含量变化的影响。设计:随机对照实验研究。地点和材料:实验地点:解放军第一军医大学中医系实验室和附属珠江医院神经内科实验室。取学习记忆合格大鼠(n=40),雌雄各半,体质量(280±30)g,随机分为空白对照组(n=8)、乌龙丹高剂量组(n=8)、低剂量组(n=8)、喜得镇组(n=8)、模型组(n=8)。干预:分离大鼠一侧颈内动脉,推注1∶200比例的干血悬液0.8mL造模。氨基酸测定用反相高效液相色谱法,乌龙丹的制剂采用水煮醇提法,灌胃给药。主要观察指标:各组大鼠Y型迷宫学习测试和脑组织20种氨基酸含量的变化。结果:模型组大鼠躲避电击错误次数最多;乌龙丹给药组的错误次数明显减少(P<0.05);用药组和对照组自身对照,造模前后大鼠的学习记忆水平有显著性差异(P<0.05);大鼠造模后,脑组织中兴奋性氨基酸天冬氨酸、谷氨酸的含量明显降低;而乌龙丹高剂量组的谷氨酸含量与模型组有显著性差异(F=3.672,P<0.05);大鼠造模后脑组织中抑制性氨基酸γ-氨基丁酸、组氨酸、牛氨酸含量明显降低,乌龙丹高剂量组γ-氨基丁酸、牛氨酸、甘氨酸,低剂量组

Abstract:BACKGROUND: Wulong dan improves learning and memory function obstacle of rats with multi-infarct dementia(MID) . Is the mechanism related with the decrease of the contents of some amino acids in brain tissue?OBJECTIVE: To investigate the effect of wulong dan on learning and memory function and contents variation of twenty amino acids in the brain tissue of model rats with MID.DESIGN: A randomized controlled experimental research.SETTING and MATERIALS: Experiment setting: Laboratory of the Departinent of Traditional Chinese Medicine, First Military Medical University of Chinese PLA; Laboratory of the Department of Neurology, Affiliated Zhujiang Hospital of the First Military Medical University of Chinese PLA. Rats with qualified learning and memory function( n = 40), half female and half male,weighing(280 ± 30) g, were allocated randomly into empty controlled group ( n = 8), high-dose wulong dan group( n = 8), low-dose wulong dan group ( n = 8), Xidezhen group( n = 8), and model group( n = 8).INTERVENTIONS: The internal cervical artery on one side of the rat was separated. The model was made by injecting 0. 8 mL dry blood suspension with the ratio of 1: 200. The content of amino acid was measured by reversed-phase high-performance liquid chromatography. Boiling and alcohol abstracting made decoction of w ulong dan. The decoction was given by intragastric administration.MAIN OUTCOME MEASURES: Y-shape labyrinth learning test results and contents variation of twenty amino acids in the brain tissues of rats in every group.RESULTS: Rats in model group made more mistakes than the others when escaping the electric shock. Rats in wulong dan group made significantly less mistakes( P < 0.05). Compared with medicine group and controlled group,there was significant difference in the level of learning and memory function of rats before and after modeling( P < 0. 05); After modeling, the content of excited amino acids in brain tissue such as aspartic acid (ASP) and glutamic acid(GLU) remakedly decreased. Significant difference was observed in the content of glutamic acid(GLU) in high dose of Wulong dan group compared with the model group( F =3. 672, P < 0. 05); After modeling, the contents of inhibited amino acids in brain tissue such as r-aminobutyric acid(GABA),hisdine(HIS) and taurine(TAU) obviously decreased. Significant difference was observed in the contents of r-aminobutyric acid(GABA), taurine (TAU) and(GLY) in high dose of wulong dan group and the content of tanrine (TAU) in low dose of wulong dan compared with the model group ( F = 3. 587,2. 695, 3. 894, 3. 124, P < 0. 05); After modeling, the contents of aromatic amino acids in brain tissue such as tyrosine(TYR) and phenylalanine(PHE) obviously decreased. Significant difference was observed in the content of tyrosine(TYR) in high dose of wulong dan group compared with the model group( F = 2. 953, P < 0. 05); After the modeling,the contents of neutrality amino acids in brain tissue such as iaoleucine(ILE),leucine(LEU), glutamine(GLN), citrulline(CIT), omiphine(ORN), lysine (LYS) obviously decreased. Significant difference was observed in the contents of leucine(LEU), glutamine(GLN), citrulline(CIT), omiphine(ORN), lysine (LYS) and valine(VAL) in high dose of wulong dan group compared with the model group( F =3. 254,2. 962,3. 417,3. 219,2. 871,3.658, P <0.05).CONCLUSION: Wulong dan can improve the learning and memory function of model rats with MID. The mechanism probably relates to the antagonizing decrease of contents of amino acids, especially the decrease of contents of excited amino acids.
