代谢组学方法研究中药穴位贴敷透皮给药治疗豚鼠哮喘的作用

目的:运用代谢组学方法研究中药"冬病夏治"方穴位贴敷法治疗哮喘的作用。方法:分组给药后取豚鼠血清,通过超高效液相色谱结合高分辨率质谱联用(UPLC-Q-TOF/MS)技术对各组血清进行代谢谱检测。原始数据采用Micromass Markerlynx用归一化的方法进行处理。输出后利用SIMCA-P+软件进行多维统计分析,通过偏最小二乘法一判别分析(PLS-DA)进行模式识别。结果:模型组和空白组、穴位给药组与模型组样本点完全分离,且各组内样品点分别具有一定的聚类作用。结论:本研究建立了基于豚鼠血清的UPLC-Q-TOF/MS代谢组学研究方法,初步阐释了中药穴位贴敷治疗哮喘的代谢组学机制。研究结果对用现代生物科学技术解释传统中药制剂理论有一定的启发作用。

Metabonomic study of therapeutical effect of herbal acupoint-application cutaneous permeable agent to the bronchial asthma cavia cobaya

Objective: To investigate the metabonomic of therapeutical effect of herbal acupoint-application cutaneous permeable agent to the bronchial asthma cavia cobaya.Methods: Blood serum from bronchial asthma cavia cobaya was taken and ultraperformance liquid chromatography coupled with Q-TOF mass spectrometry(UPLC-Q-TOF/MS) was used to detect the metabolite profiles of the blood serum samples of every group.The primary data is pretreated by the Micromass Markerlynx software.Then the data is analyzed by pattern recognition method which is composed of PLS-DA in SIMCA-P+.Results: the result showed that metabolite profiles of the blood serum sample had been separated obviously and had clustering character between the asthma-model Group and the blank control group,the acupoint administration group and the asthma-model Group.Conclusion: The topic established a method of metabonomic analysis for blood serum from cavia cobaya using LC/MS.The study shows the mechanism of metabolism for the therapy of asthma with Dongbing Xiazhi Formula.The results will be of great helpful to explain the theory of Chinese drugs pharmaceutics using biological science of modern time.
