HLA—B＊5101与白塞病的相关性研究

目的 :　探讨HLA -B 5 10 1等位基因与白塞病之间的相关性。方法 :　应用微量淋巴细胞毒试验检测HLA -A和HLA -B抗原。应用PCR -SSP技术对HLA -B5 1等位基因 (HLA -B 5 10 1～HLA -B 5 10 7)进行分析。 2 0名白塞病患者符合国际白塞病委员会分类诊断标准 ,同时以 30例正常人作对照组。结果 :　在白塞病患者中 ,HLA -B5 1表型频率明显高于对照组 患者为 6 0 % (12 2 0 ) ,对照组为 16 .7%(5 30 ) ],χ2 =10 .0 4 ,P <0 .0 0 1,校正P值 <0 .0 5 ,相对危险度为 8.98。没有发现其它HLA -A和HLA -B抗原与白塞病相关。在 12例HLA -B5 1阳性的患者中 ,均为等位基因HLA -B 5 10 1,5个HLA -B5 1阳性的对照亦均为等位基因HLA -B 5 10 1。结论 :　等位基因HLA -B 5 10 1与白塞病的易感性相关

Association between HLA - B * 5101 and Chinese patients with Behcet's disease

Objective:To investigate the association between allele HLA-B*5101 and Behcet's disease (BD).Methods: HLA-A and HLA-B typing was performed using peripheral blood lymphocytes by the standard US National Insititutes of Health (NIH) complement-dependent microlymphocyte toxicity assay. HLA-B51 allele genotyping (HLA-B*5101-HLA-B*5107) was performed by the PCR-sequence specific primer (SSP) method. Patients with BD were required to satisfy the International Study Group (ISG) criteria for diagnosis of BD. The control group consisted of 30 healthy people.Results: A highly significant HLA-B51 association with BD was determined (60% in patients vs 16.7% in controls). No other association of HLA type with BD was found. HLA-B51 allele genotyping revealed that all these HLA-B51 positive patients and HLA-B51 positive controls carried HLA-B*5101. Conclusion : This study supports an HLA-B*5101 immunogenetic predisposition and reveals a strong association of BD in Chinese patients with HLA-B*5101.
