RP-HPLC法同时测定北五味子中五味子甲素与五味子乙素含量

目的建立同时测定北五味子中五味子甲素与五味子乙素含量的方法。方法RP HPLC法。色谱柱为Thermo Hypersil BDS C18柱(4.6 mm×250 mm,5μm),流动相为甲醇乙腈水(体积比为45∶30∶25),流速1.0 mL.min-1,检测波长254 nm。结果以峰面积为纵坐标,以对照品溶液质量浓度(mg.L-1)为横坐标,进行线性回归:五味子甲素的回归方程为A=2.701×107ρ-1.396×104,r=0.999 9,表明五味子甲素在10.1~202 mg.L-1线性关系良好;五味子乙素的回归方程为A=2.797×107ρ+1.570×104,r=0.999 8,表明五味子乙素在12.2~244 mg.L-1线性关系良好。平均回收率分别为102.0%、100.5%,RSD均小于3.1%。结论该方法可用于北五味子药材质量控制。

Determination of contents of deoxyschizandrin and γ-schizandrin in Schisandra chinensis (Turcz.) Baill. by RP-HPLC

Objective To develop a RP-HPLC method for the determination of deoxyschizandrin and γ-schizandrin in Schisandra chinensis(Turcz.)Baill.in different areas.Methods A Thermo Hypersil BDS C_(18) column(4.6 mm×250 mm,5 μm)with the mobile phase consisted of methonal-acetonitrile-water(V∶V∶V=45∶30∶25)was used.The flow rate was 1.0 mL·min~(-1) and the UV detection wavelength was set at 254 nm.Results The calibration curve of deoxyschizandrin and γ-schizandrin was linear in the ranges of 10.1-202 mg·L~(-1)(r=0.999 9)and 12.2-244 mg·L~(-1)(r=0.999 8)respectively.The average recoveries of deoxyschizandrin and γ-schizandrin were 102.0% and 100.5% respectively with RSD less than 3.1%.Conclusions The method is sensitive,accurate,rapid and is suitable for the quality control of Schisandra chinensis(Turcz.)Baill.
