摘    要:目的:探讨非甲基化胞嘧啶-鸟嘌呤的寡核苷酸链(CpG ODN)对哮喘气道重塑和气道基质金属蛋白酶-9(MMP-9)表达的影响。方法:以卵蛋白致敏激发的小鼠慢性哮喘模型为对象,观察对照组、哮喘组、CpG ODN组及地塞米松组之间支气管壁厚度改变及气道MMP-9表达。结果:(1)CpG ODN组和地塞米松组的支气管壁厚度大于对照组而小于哮喘组(P〈0.05),而CpG ODN组和地塞米松组间则无显著差异(P〉0.05);(2)CpG ODN组和地塞米松组MMP-9表达高于对照组(P〈0.05)而小于哮喘组(P〈0.05),CpG ODN组和地塞米松组间则无显著差异(P〉0.05)。结论:慢性哮喘小鼠气道壁明显增厚,而早期行CpG ODN干预则可抑制肺内MMP-9的表达而减轻气道重塑。

Abstract:Objective To study the effects of CpG oligodeoxynuleotides(ODN) on airway remodeling and expression of matrix metalloprotease-9(MMP9) in a murine model of chronic asthma.Methods BALB/c mice were sensitized and repeatedly challenged with ovalbumin.Pathological slides prepared were stained with hematoxylin-eosin.The thickness of airway wall was measured.The expressions of MMP-9 were detected by immunohistochemistry.Results(1)The thickness of airway wall of CpG ODN and dexamethasone(DXM) treatment groups was higher than control group(P<(0.05)),while lower than asthma group(P<0.05).But it had not statistically significant difference between CpG ODN group and DXM treatment group(P>0.05).(2)The expressions of MMP-9 in the CpG ODN and DXM treatment groups were higher than in control group(P<0.05),while lower than in asthma group(P<0.05).It had not significant difference between CpG ODN group and DXM treatment group(P>0.05).Conclusion Airway remodeling can occur in the chronic asthma mice.Early intervention with steroid or CpG ODN may partially inhibit the expressions of MMP-9 in the lungs of chronic asthma mice,and airway remodelings are abated.
