| 摘 要： | 目的检测体外培养神经干细胞（NSC）神经生长因子（NGF），脑源性深营养因子（BDNF），神经营养因子3（NT-3）及其受体trkA、trkB、trkC的表达．方法胎鼠海马神经干细胞培养并鉴定后，提取其总RNA，RT-PCR技术检测NGF、BDNF、NT-3及其受体trkA、trkB、trkC的表达情况．免疫组化染色观察i种生长因子在NSC的分布和亚细胞定位．结果RT-PCR显示NSC表达i种生长因子及其受体mRNA．免疫组化染色显示NSC表达i种生长因子蛋白，免疫阳性产物主要分布于细胞质．结论体外培养NSC表达NGF，BDNF，NT-3及其受体，提示三种因子对NSC发育和发挥生物学功能可能有重要作用．

| Abstract: | Objective To investigate the expression of NGF,BDNF,NT-3 and their receptors trkA,trkB,trkC in cultured Neural stem cells(NSCs).Methods NSCs were isolated from embryonic hippocampus of mice.They were then cultured in vitro and identified by immunohistochemistry using nestin antibody.The expression of mRNA and protein for NGF,BDNF,NT-3 was detected by RT-PCR and immunohistochemistry.Results Significant levels of mRNA were detected for all three factors and their receptor trkA,trkB,trkC by RT-PCR.The proteins... |
