人结肠癌裸鼠移植瘤免疫组织化学与生物学特性初步观察

摘    要:目的为实验药理学开展内分泌治疗建立人结肠癌裸鼠移植瘤。方法将雌孕激素受体阳性人结肠癌移植于裸鼠并传5代,连续观察各代肿瘤病理特征、免疫组织化学、细胞动力学、DNA倍体及受体表达规律,并与原人体结肠癌比较。部分移植瘤进行超微结构透射电镜观察。结果人结肠癌裸鼠异种移植建株成功,保持原人体结肠癌相同高分化管状腺癌,持续阳性雌孕激素受体表达(生化DCC法),稳定细胞动力学特征与DNA倍体(异倍体),原代和第1、3、5代荷瘤分别显示出大肠癌最具有特征的超微结构如微绒毛和小根等,特异性大肠癌标记物癌胚抗原与花生凝集素受体表达阳性。结论性激素受体作用失常参与大肠癌发病,为内分泌治疗提供基础。

Abstract:PURPOSE In order to establish the role of endocrinal treatment in nude mice xenograft with human colonic carcinoma.METHODS A colonic tubular adenocarcinoma from a 75-year old woman was heterotransplanted to BALB/C nu/nu nude mice and maintained for five generations. The success rate of transplantation ranged between 50% and 100%. Besides the studies of pathology, cell cycle kinetics,electron microscopy, and immunohistochemistry, we also determined DNA content by FCM,ER and PR with DCC method on carcinomas from every generation on nude mice xenograft and compared the ploid status, DNA content,and the distribution of ER and PR with the original human colonic carcinoma.RESULTS Our preclinical screening indicates that the xenograft carcinomas in every generation of nude mice maintain almost the same DNA contents (mostly aneuploids),positive results of ER and PR, and the stable cell cycle features as the original human carcinoma.CONCLUSIONS This investigation could be beneficial to understand the possible mechanism of hormone receptor system imbalance as initiator and promotor for the development of some colorectal carcinomas considered as hormone-dependent tumors and provide a model to work out the genuine value of endocrinotherapy with tamoxifen prospectively for those colorectal carcinomas in the future.
