摘    要:目的 探讨不同温度对人舌鳞癌细胞系(Tca—8113)表皮生长因子受体复合物内化的影响。方法 用氯胺T法标记^125I—hEGF。采用放射受体结合分析法比较在37℃和4℃时,^125I—hEGF细胞表面结合的量和内化的量。结果 37℃时,在10min后受体复合物内化超过细胞表面结合的量。4℃时,内化量一直维持较低水平。结论 4℃时,配体结合反应稳定,适宜配体结合分析实验。EGFR复合物内化现象的研究,为临床筛选EGFR单克隆抗体进行治疗提供了实验依据。

Abstract:OBJECTIVE: To investigate the effect of different temperatures on internalization of epidermal growth factor receptor complexes in human tongue squamous carcinoma cells(Tca-8113). METHODS: Human epidermal growth factor r (hEGF) was labeled with 125I according to Chloramine-T method. The quantity of surface binding and internalization of 125I-hEGF in Tca-8113 cells at 4 degrees C and at 37 degrees C was compared respectively with radiobinding assay. RESULTS: At 37 degrees C, the amount of EGFR complexes internalization exceeded that of surface binding after 10 min. At 4 degrees C, the level of interlization maintained very low. CONCLUSION: The study indicated that the surface binding of 125I-hEGF at 4 degrees C was steadier than that at 37 degrees C. The study on phenomenon of EGFR complexes internalization provide the experimental basis for selecting anti-EGFR monoclonal antibody in clinic.
