摘    要:目的:研究舌癌exosomes诱导CTL体外抗肿瘤效应。方法:通过连续离心及超滤法从Tca8113细胞培养上清中分离、纯化出exosomes,然后与树突状细胞(DC)共培养,诱导T淋巴细胞活化为CTL。通过乳酸脱氢酶法测定特异性CTL对Tca8113的杀伤效应。采用SPSS11.0软件包对数据进行t检验。结果:exosomes可促使DC成熟。杀伤实验表明:负载exosomes的DC诱导CTL对Tca8113细胞的杀伤率显著高于未用exosomes负载的DC诱导的CTL及加IL-2培养的T细胞,其差异有显著性(P〈0.01)。结论:Tca8113细胞来源的exosomes负载DC后,可以诱导T细胞成为抗原特异性CTL,具有特异性杀伤肿瘤细胞的功能。

Abstract:PURPOSE: To investigate the anti-tumor efficacy in vitro of cytotoxic T lymphocyte(CTL) induced by the exosomes derived from tongue cancer cell line. METHODS: Exosomes was isolated and purified from the cultured supernatant of Tca 8113 by ultraspeed-centrifugation and density gradient centrifugation, and then was co-cultured with dendritic cell (DC).When DC and T cell were mix-cultured, T cell was activated into CTL(CD3^+CD8^+cells), and the shift of CD3^+CD8^+cells were analyzed by FCM. Then the antitumor efficacy of the CTL activated by DC loaded with exosomes was detected by lactate dehydrogenase (LDH) kit, according to the instruction. The data was processed with SPSS1 1.0 package for Student's t test. RESULTS: The CTL induced by DC pulsed with exosomes had significant activity of killing Tca8113 compared with CTL induced by DC without exosomes and T cells cultured with IL-2(P〈0.01). CONCLUSIONS: When loaded with exosomes derived from tongue cancer cell line, DC could activate T cells into CTL (CD3^+CD8^+T cells). The exosomes-specific CTL could kill Tca8113 cells.
