脑缺血后海马CA1区细胞凋亡现象的观察

摘    要:观察大鼠完全性前脑缺血再灌注损伤过程中海马CA1区神经元死亡的另一种形式-细胞凋亡。采用琼酯糖凝胶电泳及原位缺口翻译法,观察海马CA1区神经元是否有凋亡特征性改变。结果;脑缺血损伤后5d,从海马CA1区神经元提取DNA,其琼酯糖凝胶电泳呈现典型梯状结构。而采用原位缺口翻译法,发现缺血损伤后3d,海马CA1区开始出现胞核染色体阳性的凋亡细胞,缺血损伤后5d,凋亡细胞达到高峰。结论:海马CA1区尽管

Abstract:Objective: To observe another form of cell death (apoptosis) in the rat hippocampus CA1 pyramidal neurons following forebrain ischemia. Methods: Electrophoresis in agarose gels and in situ nick translation were employed to observe whether neurons in hippocampus CA1 appeared characteristic alteration of apoptosis. Results: DNA was extracted from hippocampus CA1 neurons 5 d after ischemic injury, which exhibited the characteristic ladder pattern in agarose gels. Using INT, positive staining was occasionally seen in nuclei of the hippocampus CA1 neurons 3 d after ischemic injury; apoptotic neurons reached a maximum 5 d after ischemic injury. Conclusions: Apoptotic mechanism may play an important role in the delayed death of hippocampus CA1 neurons. It may provide novel therapeutic approaches for prevention of cerebral ischemic injury.
