摘    要:目的:建立以反相高效液相色谱法同时测定地锦草药材中4种黄酮类成分含量的方法。方法:色谱柱为Hypersil BDS C18(150mm×4.6mm,5μm),流动相为甲醇-0.3%磷酸(梯度洗脱),流速为1.0mL·min-1,柱温为30℃,检测波长为360nm。结果:地锦草中芦丁、槲皮素、木犀草素和山柰酚4种黄酮类成分的进样量分别在0.1272~2.0344μg(r=0.9999)、0.0891~1.4256μg(r=0.9998)、0.0766~1.2264μg(r=0.9997)、0.1130~1.8080μg(r=0.9998)范围内与各自峰面积积分值呈良好的线性关系;四者平均回收率分别为97.6%、99.4%、98.1%、101.9%,RSD分别为0.94%、0.73%、0.99%、1.34%(n均=9)。结论:本方法操作方便、结果准确、重现性好,可用于地锦草药材的质量控制。

Abstract:OBJECTIVE: To establish a RP- HPLC method for simultaneous determination of 4 flavonoids in Herba Euphorbiae Humifusae.METHODS: The samples were separated on Hypersil BDS C18 column(150 mm × 4.6 mm, 5μm) with CH3OH-0.3%HaPO4 as mobile phase(gradient elution) at a flow rate of 1.0 mL · min^-1.The column temperature was maintained at 30℃ and the detective wavelength was set at 360 nm.RESULTS: The linear ranges of 4 flavonoids including rutin, quercetin, luteolin and kaempferol were 0.127 2-2.034 4 μg(r = 0.999 9), 0.089 1-1.425 6 μg(r=0.999 8), 0.076 6-1.226 4μg(r = 0.999 7), 0.113 0-1.808 0 μg(r = 0.999 8) respectively, and their average recoveries were 97.6% , 99.4%, 98.1% and 101.9%, respectively with RSD at 0.94%, 0.73%, 0.99% and 1.34%, respectively (all n = 9) . CONCLUSION: This method was proved to be simple, accurate, reproducible and applicable for the quality control of Herba Euphorbiae Humifusae.
