摘    要:目的:建立以高效液相色谱法测定皂角刺提取物中槲皮素含量的方法。方法:色谱柱为Dikma Kromasil C18(250mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸溶液(30∶70),检测波长为360nm,柱温为30℃,流速为1.0mL·min-1。结果:槲皮素进样量在0.134~1.072μg范围内与峰面积积分值呈良好的线性关系(r=0.9999);平均回收率为98.98%,RSD=1.87%(n=9)。结论:本方法灵敏、准确、简便、快速,可用于皂角刺提取物的质量控制。

Determination of Quercetin in Extracts of Gleditsia sinensis by HPLC

Abstract:OBJECTIVE: To establish an HPLC method for the determination of quercetin in the extracts of Gleditsia sinensis .METHODS: The chromatographic separation was performed on Kromasil C18 column(250mm×4.6mm,5μm) with column temperature kept at 30 ℃ .The mobile phase consisted of acetonitrile - 0.1% phosphoric acid solution(30 : 70) at a flow rate of 1.0 mL · min^-1 with detection wavelength set at 360 nm. RESULTS: The linear range of quercetin was 0.134--1.072 μg(r = 0.999 9) with its average recovery at 98.98% (RSD = 1.87%, n = 9) . CONCLUSION: The established method was proved to sensitive, accurate, simple, rapid and applicable for the quality control of the extracts of Gleditsia sinensis.
