盐酸二甲双胍对非酒精性脂肪肝大鼠脂肪组织相关基因表达的影响

摘    要:目的:从基因表达水平上分析盐酸二甲双胍对非酒精性脂肪肝(NAFLD)大鼠模型的干预效果及其作用机制。方法:以高脂饲料饲喂雄性Wistar大鼠诱发NAFLD,并分别进行盐酸二甲双胍干预与蒸馏水灌胃对照,每组8只,为期4周。通过逆转录和定量聚合酶链式反应(RT-qPCR)方法检测脂肪组织中脂联素、瘦素、肿瘤坏死因子(TNF)-α、抵抗素和解偶联蛋白2的mRNA相对水平并进行t检验。结果:与对照组比较,药物干预组脂联素mRNA相对水平略有上升(P>0.05),瘦素mRNA和TNF-αmRNA相对水平显著下降(P<0.05)。2组样品中均未检测到抵抗素和解偶联蛋白2的mRNA。结论:盐酸二甲双胍可能主要是通过促进脂联素基因表达和抑制TNF-α基因表达来调节肝脏脂肪代谢、改善肝细胞脂肪变性和炎症反应,从而发挥其对NAFLD的干预作用。

Effect of Metformin Hydrychloride on the Related Gene Expression in Adipose Tissue of Rats with Non- alcoholic Fatty Liver Disease

Abstract:OBJECTIVE: To study the interventional effect and mechanism of metformin hydrychloride on gene expressions in rats with non-alcoholic fatty liver disease (NAFLD) . METHODS: The male Wistar rats were fed with high fatty chow to induce NAFLD. And then these rats were fed with metformin hydrychloride (intervention group, n = 8) or distill water (control, n = 8) for 4 weeks. The relative levels mRNA of adiponectin, leptin, tumor necrosis factor a (TNF-α), resistin and uncoupling protein 2 (UCP2) in the adipose tissue was detected by RT- qPCR. And the data were subjected to a t test. RESULTS: As compared with the control group, the relative level of mRNA of adiponectin in the intervention group was slightly up- regulated (P 〉 0.05), but the relative levels of the mRNA of TNF-α and leptin were significantly down-regulated (P〈0.05) . Neither resistin mRNA nor uncoupling protein 2 mRNA was detected in the two groups. CONCLUSION: The interventional effect of metformin hydrychloride on the NAFLD rats were possibly achieved by regulating fat metabolism and improving adipose degeneration and inflammatory reaction in liver cells through promoting gene expression of adiponectin while inhibiting the gene expression of TNF-α.
