脂联素在非小细胞肺癌组织中的表达及其与基质金属蛋白酶-9 和肿瘤血管生成的关系

摘    要:目的:探讨脂联素在人非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中的表达及其与基质金属蛋 白酶(matrix metalloproteinase,MMP)-9和肿瘤血管生成的关系。方法:免疫组织化学法检测87例人NSCLC及10例正常 肺组织中脂联素和MMP-9的表达及其微血管密度(microvessel density,MVD)。结果:NSCLC组织中脂联素阳性表达 率(49.43%)明显低于正常肺组织(90.00%,P<0.05),MMP-9的阳性率(59.77%)明显高于正常肺组织(10.00%,P<0.05), NSCLC中MVD值(14.58±0.67)明显高于正常肺组织中的MVD值(0.80±0.12),P<0.05];脂联素的阳性表达率在不同的肿 瘤大小及临床分期组间及是否淋巴转移组间差异有统计学意义(P<0.05);MMP-9阳性表达率在不同的患者年龄、大 小、临床分期组间及是否淋巴转移组间差异有统计学意义(P<0.05),淋巴转移组的MVD值明显高于无淋巴转移的MVD 值(P<0.05);脂联素表达与MMP-9表达、MVD值呈负相关(r值分别为?0.55,?0.22;P<0.05),而MMP-9表达与MVD值呈 正相关(r=0.21,P<0.05)。结论:NSCLC组织中脂联素低表达,可能负性调控MMP-9表达从而参与NSCLC的发生发展。

Abstract:Objective: To explore the expression of adiponectin and its relationship with matrix metalloproteinase (MMP)-9 and angiogenesis in non-small cell lung cancer (NSCLC) tissue. Methods: A total of 87 samples of NSCLC and 10 samples of normal lung tissue were collected for detection of adiponectin and MMP-9 expression by immunohistochemical method. Anti-CD34 monoclonal antibody against endothelial marker was used to evaluate microvessel density (MVD). Results: Th e positive rate for adiponectin in NSCLC (49.43%) was signifi cantly lower than that in normal lung tissues (90.00%) while for MMP-9 in NSCLC (59.77%) was significantly higher than that in normal lung tissues (10.00%, P<0.05); MVD in NSCLC (14.58±0.67) was higher than that in normal lung tissues (0.80±0.12), P<0.05]. In NSCLC, the positive rates of adiponectin and MMP- 9 were varied in groups with different lymph node metastasis, tumor size and clinical stage (P<0.05). Meanwhile, MVD value in NSCLC with or without lymph node metastasis was significantly different. There was a negative correlation between adiponectin level and CD34 (MVD) or MMP-9 level (r=?0.22 or r=?0.55, respectively, P<0.05). On the contrary, MMP-9 was positively correlated with MVD (r=0.21, P<0.05). Conclusion: The decreased expression of adiponectin might be related to the development of NSCLC through negative regulation of MMP-9 expression.
