摘    要:目的:探讨高度近视眼视网膜-脉络膜-巩膜血管活性肠肽(vasoactiveintestinalpeptide,VIP)分子的动态表达及意义。方法:选择出生1d龄黄鸡共42只,其中21只小鸡右眼遮盖作为实验组,分别持续遮盖1,2,4wk,每周7只小鸡。另21只小鸡双眼均未遮盖作为对照组。两组均采用检影验光检测屈光度;眼科A超测定新鲜离体眼轴长度;对两组3个时间段后部眼球壁组织行SP法免疫组织化学染色检测VIP的动态表达。结果:实验组由实验前远视眼快速演变为高度近视眼,并随遮盖时间延长,近视度数加深,眼轴延长。两组视网膜、脉络膜均有VIP表达:广泛表达于视网膜内核层、内网状层和脉络膜;在神经节细胞层和神经纤维层也有弱表达;强阳性表达位于视网膜光感受器外节。随出生时间延长两组VIP表达均上调。与对照组相比,实验组VIP表达明显上调(P<0.01)。结论:形觉剥夺可导致高度近视眼;VIP表达主要位于视网膜光感受器外节、内核层、内网状层和脉络膜,神经节细胞层和神经纤维层也有弱表达;高度近视眼较正常眼VIP表达明显上调。

Abstract:AIM: To investigate the dynamic expression of vasoactive intestinal peptide on retina-choroids-sclera in high myopia. METHODS: 42 yellow chickens of 1d old were used for research. The experimental group included 21 chickens'right eyes which were covered as form deprivation for 1,2 and 4wk respectively. The other 21 chickens were not covered as normal control group. In both groups, diopter was detected by retinoscopic refraction, eyeball axis was determined by Ultrasound-A, VIP expression on retina-choroids-sclera at three stages was examined by SP immunohistochemistry staining. RESULTS: The experiment eyes were changed from hypermetropia prior experiment to high myopia during experimental stages. The diopter degrees were becoming deeper and eyeball were becoming longer accompanying with longer time of form deprivation. Both groups had the expression of VIP. Strong expression of VIP existed on photoreceptor-outer segment of retina; Widespread expression existed on inner nuclear layer and inner plexiform layer of retina and choroids; Weak expression existed on ganglion cell layer and nerve fiber layer. The expression was up-regulated following the age. Compared with the control, the expression of protein of VIP in the experimental group was siginificantly higher(P<0.01). CONCLUSION: Form deprivation can induce high myopia. The expression of VIP exists on photoreceptor- outer segment, inner nuclear layer and inner plexiform layer of retina and choroids, and weak expression exists on ganglion cell and nerve fiber layer. VIP expression is significantly up-regulated in high myopia.
