HBcAg/HBeAg对慢性乙型肝炎患者PBMC中Th1/Th2类细胞应答的影响

目的 探讨ＨＢｃＡｇ／ＨＢｅＡｇ对慢性乙型肝炎患者ＰＢＭＣ中Ｔｈ１／Ｔｈ２类细胞应答的影响。方法 用套式ＰＣＲ法检测６４便慢性ＨＢＶ感染者ＰＢＭＣ中ＨＶＢ ＤＮＡ；分别用ＰＨＡ、ＨＢｃＡｇ和ＨＢｅＡｇ体外培养；ＥＬＩＳＡ法检测ＰＢＭＣ产生Ｔｈ１类细胞因子（ＩＬ－２、ＩＦＮ－γ）和Ｔｈ２类细胞因子（ＩＬ－４、ＩＬ－１０）的含量。结果 表明ＨＢＶ ＤＮＡ阳性组和阴性组相比，无论是在ＰＨＡ还是在ＨＢｃＡ

The effect of HBcAg/HBeAg on Th1/Th2 response in PBMC of chronic HBV infection patients

Objective To explore the effect of HBcAg/HBeAg on Th1/Th2 response in PBMC of chronic HBV infection patients. Methods HBV DNA in PBMC of 64 cases with chronic HBV infection were detected by nested polymerase chain re- action. Thl-like cytokines (IL-2,IFN-γ) and Th2-like cytokines (IL-4,IL-10),produced by PBMC stimulated by PHA、 HBcAg、 HBeAg respectively,were measured by ELISA. Results There were no significant difference of both the levels of Th1- like cytokines and the levels of Th2-like cytokines between HBV DNA positive group and HBV DNA negative group when PBMCs were induced by PHA、HBcAg、HBeAg respectively. HBcAg-induced group and HBeAg-induced group were compared with PHA-induced group,the Th1-like cytokines production was predominant when PBMC were stimulated by HBcAg; in contrast,Th2-like cytokines production was predominant when PBMC were stimulated by HBeAg. Conclusions The results suggested that HBVs invasion of PBMC did not affect the Thl/Th2 response; HBcAg and HBeAg could possibly affect the production of Th1/Th2-like cytokines in PBMC of chronic HBV infection cases.
