Expression and significance of IL-4, IFN-y in liver fibrosis of rats infected with Pagumogonimus skrjabini

Abstract:Objective To explore the expression and significance of interleukin-4 and interferon-y in the formation of liver fibrosis of rats infected with Pagumogonimus skrjabini, and provide experimental evidence on the role of Th1/Th2 for the formation and regulation of liver fibrosis of rats infected with Pagumogonimus skrjabini. Methods 36 rats were randomly divided into two groups, the control group and the infection group. The Infection group were divided into 6 groups according to infection time: group A (1 week after infection), group B (2 weeks after infection), group C (4 weeks after infection), group D (8 weeks after infection), group E (12 weeks after infection), F group (16 weeks after infection). Each rat was infected with 15 Pagumogonimus skrjabini metacercarias by the way of peritoneal injection. The rats were sacrificed and observed indicators in corresponding time after infection. The pathomorphological changes of liver fibrosis were observed by the HE staining and VG staining. The immunohistochemical technique was used to measure the dynamic changes of expression of interleukin-4, interferon-y in the formation of liver fibrosis in rats infected with Pagumogonimus skrjobini. Results The results of pathomorphology showed that the ratio of collagen fiber area (S/T) and the liver fibrosis rating increased and the degree of liver fibrosis gradually deepened gradually with the extension of infection time. The results of immunohistochemistry showed that the percentage of positive cells of INF-7 expression increased gradually at lst-Sth week after infection and reached the peak at the end of 8th week, then decreased gradually at 12th-16th week. The percentage of positive cells of IL-4 expression increased gradually at lst-16th week after infection and rose rapidly at the end of 12th week. Conclusion The study suggested that Pagumogonimus skrjabini infection in rats could cause liver fibrosis, the expression of IL-4 and IFN-7 increased in the formation of liver fibrosis of rats infected with Pogumogonimus skrjabini, the extent of liver fibrosis was associated with the infection time and the expression of IL-4, IFN-y. The study prompted that cytokines IFN-y secreted by Thl and IL-4 secreted by Th2 played a role in spontaneous immune regulation during the liver fibrosis formation of rats infected with Pagumogonimus skrjohini.
