摘    要:目的:优选黄独总皂苷的分离纯化工艺.方法:以黄独总皂苷含量为指标,采用紫外分光光度法测定其含量,动态吸附-解吸法筛选大孔树脂型号,并优选黄独总皂苷的分离纯化工艺.结果:D-101型大孔树脂富集纯化黄独总皂苷的最佳工艺为黄独提取液质量浓度0.2 g·mL-1,上样量6 BV,流速1.0 mL·min-1,加4 BV水洗除杂,50%乙醇4 BV解吸,收集洗脱液.此工艺条件下所得提取物黄独总皂苷质量分数61.85%.结论:该优选工艺简单可行,能满足大生产实际要求.

Abstract:Objective: To optimize separation and purification technology of total saponins from Dioscorea bulbifera. Method: With the content of total saponins from D. bulbifera as index,which was determined by ultraviolet spectrophotometry,optimum macroporous resin model was selected by dynamic adsorption-desorption method,then separation and purification technology of total saponins from D. bulbifera was optimized. Result: Optimum separation and purification technology conditions of total saponins by D-101 macroporous resin were as follows:the concentration of extraction liquid from D. bulbifera 0.2 g·mL-1,sample volume of 6 BV,flow rate of 1.0 mL·min-1, washed impurity with 4 BV water,and then desorpted with 4 BV 30% ethano1,collected eluate.Under these technology conditions,yield of total saponins from D. bulbifera reached as high as 61.85%. Conclusion: This optimized technology was simple and feasible,which could meet industrial requirements.
