黄芪注射液对高果糖饮食诱导的大鼠胰岛素抵抗和氧化应激的影响

摘    要:目的:探讨黄芪注射液对高果糖饮食大鼠胰岛素抵抗和氧化应激的抑制作用.方法:SD大鼠经高果糖饲料喂养4周后,随机分为模型组,黄芪注射液高、低剂量(相当于生药量ip 5,2.5 g·kg-1)组、罗格列酮(3 mg·kg-1,ig)组,连续给药4周,继续给予高果糖饲料4周.同时设立正常组,普通饲料喂养.测定体重、空腹血糖、血浆胰岛素、胰岛素抵抗指数、氧化应激指标.结果:模型组体重与正常组无明显区别;空腹血糖、血浆胰岛素水平、胰岛素抵抗指数、丙二醛(MDA)含量明显高于正常组(P<0.05);过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性显著低于正常组(P<0.05).与模型组相比,黄芪注射液组、罗格列酮组空腹血糖、血浆胰岛素水平、胰岛素抵抗指数、MDA含量降低,CAT,SOD,GSH-Px活性升高,高剂量组与模型组比较有显著性差异(P<0.05).结论:黄芪注射液可减轻高果糖饮食所引起的胰岛素抵抗及氧化应激.

Abstract:Objective: To investigate the depressant effect of Astragali injection on insulin resistance and oxidative stress in rats treated by high-fructose diet. Method: SD rats were randomly divided into model group, Astragali injection high, low dose (5, 2.5 g·kg-1, equivalent to the amount of crude drug, ip) groups, rosiglitazone (3 mg·kg-1, ig) group after fed with high-fructose forage for 4 weeks and were given high-fructose forage for another 4 weeks. The rats in the normal group were given ordinary forage. The body weight, fasting blood glucose, plasma insulin, insulin sensitivity index, oxidative stress indicators were determined. Result: There was no significant difference between model group's body weight and normal group's body weight; the fasting blood glucose, plasma insulin levels, insulin resistance index, (MDA) content were markedly higher than normal group (P<0.05); catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) activities were significantly lower than those in normal group (P<0.05). Compared with the model group, the fasting blood glucose, plasma insulin levels, insulin resistance index, malonaldehyde MDA content decreased, while CAT, SOD, GSH-Px activities increased in the Astragali injection groups and rosiglitazone group. There was significant difference between high dose group and model group (P<0.05). Conclusion: Astragali injection can release insulin resistance and oxidative stress induced by high-fructose diet.
