HPLC测定防风通圣丸中2个色原酮苷类成分含量

目的:建立防风通圣丸中2个色原酮苷类成分的含量测定方法.方法:采用HPLC法,对防风通圣丸中的升麻素苷、5-O-甲基维斯阿米醇苷进行定量分析.kromasil C18柱(4.6 mm×250 mm,5μm)；流动相甲醇-乙腈-0.1％磷酸(24:12:64)；检测波长254 nm.结果:升麻素苷、5-O-甲基维斯阿米醇苷分别在0.0125 ～0.250,0.0108 ～0.216 μg(r ＞0.9999)线性关系良好,平均回收率＞96.75％,RSD＜ 1.65％.结论:该研究同时进行升麻素苷、5-O-甲基维斯阿米醇苷的定量分析,可作为防风通圣丸的质量控制标准.

Determination of Two Chromone Glycosides in Fangfeng Tongsheng Wan by HPLC

Objective:To develop RP-HPLC method for the quantitative determination of two chromones in Fangfeng Tongsheng Wan.Method:The determination was performed on a kromasil C18 column(4.6 mm×250 mm,5 μm) with methanol-acetonitrile-0.1% phosphoric acid(24 ∶ 12 ∶ 64) as the mobile phase at the flow rate of 1.0 mL·min-1.The detection wavelength was set at 254 nm.Result:The calibration curves of prim-O-glucosylcimifugin and 4′-O-glucosyl-5-O-methylvisamminol were linear in the ranges of 0.012 5-0.250 μg and 0.010 8-0.216 μg(r > 0.999 9),respectively.The average recoveries were over 96.75% with RSD lower 1.65 %.Conclusion:The method is simple,accurate,reproducible,and suitable for simultaneously determination of prim-O-glucosylcimifugin and 4′-O-glucosyl-5-O-methylvisamminol,and can be used for quality control of Fangfeng Tongsheng Wan.
