土贝母苷甲对人肺腺癌细胞增殖及凋亡的影响

目的探讨土贝母苷甲对人肺腺癌细胞(A549)增殖及凋亡的影响。方法应用MTS法检测土贝母苷甲不同浓度、不同时间对A549细胞增殖的影响;使用1μg/mL土贝母苷甲处理A549细胞48 h作为处理组,对照组加入培养基培养48h;使用流式细胞仪分别检测两组细胞的周期分布及细胞凋亡率;并使用Western blot检测两组细胞凋亡相关通路Caspase蛋白的表达。结果MTS实验结果显示各组细胞随着土贝母苷甲浓度的增加A549细胞的增殖速度降低,并在培养48h后,各组细胞的增殖能力出现显著差异(P<0.05);流式细胞仪检测土贝母苷甲处理A549细胞48 h后比对照组细胞凋亡率显著增高(P<0.05);处理组A549细胞更多的阻滞于G0/G1期(P<0.05);Western blot检测各组细胞Caspase通路蛋白表达显示处理组比对照组Caspase-9、Caspase-3和Parp蛋白表达增加(P<0.05)。结论土贝母苷甲可抑制人肺腺癌细胞增殖,并诱导人肺腺癌细胞凋亡,具有一定的抗肿瘤作用,为中医药土贝母在临床应用中奠定基础。

Effect of tubeimoside-1 on proliferation and apoptosis of human lung adenocarcinoma cells

Objective To investigate the effect of tubeimoside-1 on the proliferation and apoptosis of human lung adenocarcinoma cells(A549).Methods MTS method was used to detect the effects of different concentrations and time of tubeimoside-1 on cell proliferation of A549 cells.A549 cells were treated with 1ug/ml tubeimoside-1 for 48 h as the treatment group,and the control group was added with culture medium for 48 h.Cytometry was used to detect the cell cycle distribution and apoptosis rate of the two groups.Western blot was used to detect the expression of Caspase protein in the apoptosis-related pathways of the two groups.Results The results of MTS experiments showed that the cell proliferation rate of A549 cells decreased with the increase of the concentration of tubeimoside-1,and after 48 hours of culture,the cell proliferation ability of each group showed a significant difference(P<0.05).Flow cytometry apoptosis rate of A549 cells treated with tubeimoside-1 for 48 h was significantly higher than that of the control group by cytometry.A549 cells in the treatment group were more arrested in the G0/G1 phase(P<0.05).Caspase blot pathway protein expression was detected by Western,and it showed that the expression of Caspase-9,Caspase-3 and Parp protein in the treatment group was increased compared with the control group(P<0.05).Conclusion Tubeimoside-1 can inhibit the proliferation of human lung adenocarcinoma cells and induce apoptosis of human lung adenocarcinoma cells.It has a certain antitumor effect,which lays the foundation for the clinical application of traditional Chinese medicine.
