大鼠局灶脑缺血和(或)再灌流模型中IL-1β的表达及其与白细胞浸润的关系

摘    要:目的 探讨白细胞介素 1(IL 1)在脑缺血和 (或 )再灌流中的动态变化及其与白细胞浸润的关系。方法 采用大鼠大脑中动脉梗塞模型 ,分别用免疫组织化学方法检测IL 1β、酶组化方法检测髓过氧化物酶 (MPO)的动态变化 ,用图象分析进行定量分析。结果 显示缺血 3h及再灌流 0 .5hIL 1β表达增高 (P <0 .0 5 )并逐渐增高 ,2 4h达高峰 (P <0 .0 1) ,在持续缺血 12 0h仍有意义 ,而再灌流 12 0hIL 1β恢复至缺血前水平 (P >0 .0 5 )。缺血组和缺血再灌流组IL 1β与MPO水平呈非常显著正相关 (P <0 .0 1)。结论 提示缺血脑区IL 1β是由局部神经元、星形细胞和血管内皮细胞合成 ,其介导的缺血再灌流损伤与白细胞浸润有密切关系 ,说明在脑缺血和 (或 )再灌流损伤中 ,高浓度的IL 1β是一个重要因素。

Abstract:Objective to Know the dynamic changes of interluekin-1(IL-1)in cerebral ischemia/reperfusion and its relation with leukocyte infiltration Method A model of rat middle cerebral artery was used to detect the dynamic changes of IL-1 β with immunohistochemical method and those of myeloperoxidase (MPO) with zymohistochemical method respectively A quantitative analysis was made with an image pattern analysis Result It was found that the expression of IL-1 β increased at 3 h of the ischemia group (I) and at 0.5 h of the ischemia/reperfusion group (IR), arrived at the peak after 24 h and remained at a significantly high level on the 120 h of ischemia ( P <0.01) On the 120 h of reperfusion the expression of IL-1 β returned to the preischemic level ( P >0.05) MPO showed a clear increase in ischemia group at 3 h and ischemia/reperfusion group 0.5 h and peak time appeared at 24 h ( P <0.01); The IL-1 β in ischemic group and ischemic/reperfusion group was very coherent with MPO ( P <0.01) Conclusion These finding suggest that IL-1 β be most likely synthesized in the neurons, astrocytes and vascular endothlial cells of the ischemic region of the brain and the mediated damages of ischemia/reperfusion be closely related to the leucocytic infiltration So, it is concluded that a high concentration of IL-1 β is one of the important factors to result in damages in cerebral ischemia/reperfusion
