小柴胡汤诱导荷瘤小鼠S_(180)细胞凋亡及对细胞周期的影响

摘    要:目的探讨小柴胡汤SRBD对荷瘤小鼠S180的抑瘤作用及可能机制。方法通过体内实验,观察不同浓度SRBD对小鼠肉瘤S180的抑制作用,用流式细胞仪(FCM)检测法研究SRBD诱导肿瘤细胞凋亡以及对肿瘤细胞周期的影响。结果SRBD中、小剂量组中SRBD对荷瘤鼠S180细胞有明显抑制作用,各剂量组SRBD均可诱导荷瘤鼠S180细胞凋亡,与对照组比较有显著性差异(P<0.05);SRBD中、小剂量组S期细胞数明显减少(P<0.05),G0/G1期细胞数明显增加(P<0.05),S180细胞增殖指数明显下降(P<0.05)。结论SRBD对小鼠肉瘤S180的生长具有抑制作用,其机制可能与诱导肿瘤细胞凋亡及抑制癌细胞增殖周期有关。

Abstract:Objective It is to discuss the anti-tumor function of small Radix Buppleuri decoction (SRBD) on bearing cancer mice S180 and its possible mechanism. Methods The inhibition of different concentration SRBD on sarcoma180 was observed with in vivo experiment. The apoptosis of tumor cells induced by SRBD and its influence on cell cycle were studied with flow cytometry. Results SRBD had obvious inhibition on bearing cancer mice S_(180) cell in the medium dose and small dose groups. SRBD could induce the apoptosis of bearing cancer mice S_(180) cell in every group, the difference was significant compared with control group (P<0.05). In the medium dose and small dose groups, the cells quantity in S stage decreased obviously (P<0.05) and that in G0/G1 stage increased obviously (P<0.05), S_(180) cell proliferation index debased obviously (P<0.05). Conclusion SRBD has anti-tumor effect on the growth of bearing cancer mice S_(180) cell, the mechanism may be related with inducing tumor cell apoptosis and inhibiting the proliferation cycle of tumor cells.
