布洛芬糖浆剂药动学及相对生物利用度

摘    要: 目的:测定布洛芬糖浆剂的药动学及相对生物利用度。方法:8名健康男性志愿者单剂量随机交叉po400mg布洛芬后,采用HPLC测定血浆药物浓度。结果:口服糖浆剂及片剂后血药浓度达峰时间分别为(0.86±0.30)h和(1.78±0.34)h,峰浓度分别为(48.9±9.71)μg·ml-1和(44.2±8.23)μg·ml-1,药 时曲线下面积分别为(175.1±31.9)μg·h·ml-1和(178.6±23.2)μg·h·ml-1。结论:统计分析表明,糖浆剂较片剂达峰时间短,吸收快。布洛芬糖浆剂相对生物利用度为(98.3±14.5)%。

Abstract:OBJECTIVE: To determine the pharmacokinetics and relative bioavailability of ibuprofen syrup. METHODS: The drug concentration in plasma was assayed by HPLC method after administration of a single oral dose of 400 mg ibuprofen to each of 8 healthy male volunteers. RESULTS: The peak levels (tp) in plasma averaged (48.9±9.71)μg·ml-1 and (44.2±8.23)μg·ml-1 at (0.86±0.30)h and (1.78±0.34)h, and the areas under the drug concentration curves (AUC) were (175.1±31.9)μg·h·ml-1 and (178.6±23.2)μg·h·ml-1 for ibuprofen syrup and tablets, respectively. CONCLUSION: The result of statistical analysis showed that the syrup could reach peak level faster than tablets.
