乌头多糖的提取分离与含量测定

摘    要:目的:从中药乌头(Aconitum Paniculigerum Nakai)中提取多糖并对其进行含量测定。方法:乙醇超声提取得到多糖,采用蒽酮-硫酸比色法、紫外可见分光光度法对其进行含量测定,检测波长为(490±1)nm。结果:乌头多糖在0.020 2~0.101 0 mg/ml范围内有良好的线性关系,r=0.999 7,平均回收率为100.60%,RSD=1.97%。药材中多糖以葡萄糖为标准含量,含量为15.82%。结论:该方法准确快速、重现性好。

Abstract:Objective: To extract and determine the content of polysaccharide in Aconitum Paniculigerum Nakai.Methods: Polysaccharide was abtained by ethanol,and the content of polysaccharide was identified by anthrone-sulfuricacid colorimetry and UV method with the wavelength was(490±1) nm.Results: Polysaccharide had a good linearity within 0.020 2-0.101 0 mg/ml(r=0.999 7),the average recovery was 100.60% and RSD was1.97%.Conclusion: This method is quick,accurate and reliable for determination of Aconitum Paniculigerum Nakai.
