| 摘 要： | 在卡介苗加脂多糖（ＢＣＧ＋ＬＰＳ）小鼠肝损伤模型，激活或封闭肝巨噬细胞，测定血浆肿瘤坏死因子（ＴＮＦ），谷丙转氨酶（ＧＰＴ），谷草转氨酶（ＧＯＴ）的变化并检查肝组织的病理改变．注射ＢＣＧ后，再注射微量ＬＰＳ，小鼠出现血浆ＴＮＦ，ＧＰＴ，ＧＯＴ剧烈升高及严重的肝损伤．以维生素Ａ激活肝巨噬细胞，ＢＣＧ＋ＬＰＳ诱导的肝损伤加剧．以印度墨汁或硅砂封闭肝巨噬细胞，则ＢＣＧ＋ＬＰＳ诱导的肝损伤减轻．可见，ＢＣＧ＋ＬＰＳ诱导的小鼠肝损伤与肝巨噬细胞的功能关系密切，来源于肝巨噬细胞的ＴＮＦ在ＢＣＧ＋ＬＰＳ诱导的肝损伤中起重要作用．

| Abstract: | To determine the possible involvement oftumor necrosis factor-α (TNF) in Bacillus Calmétte Guérin (BCG) + lipopolysaccharide (LPS)-induced liver damage and its relation to hepatic macrophages(HM Φ ), the influences of stimulation and blockade of HM Φ on this model were studied. Injection of LPS following the BCG injection resulted in a remarkable elevation of circulating TNF, glutamic pyruvic transaminase, glutamic oxaloacetic transaminase levels, and severe liver damage. These pathological alterations were enhanced by the activation of HM Φ with retinol and reduced by the blockade of HM Φ with silica and Indian ink. These findings suggest that the degree of liver injury induced by BCG+LPS is closely related with HM Φ function, and that TNF plays an important role in the pathogenesis of the liver injury. |
