红珍珠血脂康胶囊的降血脂药效学研究

摘    要:背景通过红珍珠血脂康胶囊药效学研究,使青藏高原特色的唐古特白刺等天然植物资源得到可持续利用与开发.目的探讨以青藏高原和柴达木地区丰富的唐古特白刺、沙棘和枸杞等天然植物资源的果实为主要原料开发的红珍珠血脂康胶囊调节血脂的作用机制.设计随机对照的实验研究.地点、材料和干预实验在青海省高原医学科学研究所药理实验室完成.昆明种小白鼠 66只和 Wistar大白鼠 68只,雌雄各半.按随机数字表法将小鼠分为普通饲料喂养组 12只 (作空白对照 ),高脂饲料喂养鼠 54只,饲喂 2周后,随机分为高脂模型组,血脂康高剂量 (血脂康 2.0 g/kg)组,低剂量 (血脂康 1.0 g/kg)组,阳性对照 (脂必妥片 1.05 g/kg)组.灌胃给药 10 mL/kg,空白对照组和高脂模型组给予同体积自来水,测定小鼠血清总胆固醇和三酰甘油水平.将健康大白鼠 68只按随机数字表法分为空白对照组 (普通饲料 ),高脂模型组以及血脂康低剂量组 (0.5 g/kg)、中剂量组 (1.0 g/kg)、高剂量组 (2.0 g/kg)3个组和阳性对照组 (脂必妥片 1.05 g/kg).灌胃给药 10 mL/kg,空白对照组和高脂模型组灌胃给予同体积自来水.测定大鼠血清总胆固醇、三酰甘油、高密度脂蛋白胆固醇、丙二醛水平,红细胞超氧化物歧化酶( superoxide dismutase,SOD)活力.结果 血脂康高、低剂量组高脂血症大鼠和小鼠血清总胆固醇、三酰甘油水平明显低于高脂模型组( P《 0.05~ 0.01).血脂康高、中剂量组高脂血症大鼠血清丙二醛水平 (6.17± 1.81) ,(6.89± 1.54)μ mol/L]明显低于高脂模型组 (8.64± 2.07) μ mol/L]( P《 0.05~ 0.01);血脂康高、中、低剂量组高脂血症大鼠红细胞 SOD活力明显高于高脂模型组比( P《 0.05).结论 提示红珍珠血脂康胶囊具有预防高脂血症、改善动脉粥样硬化等作用.

Abstract:BACKGROUND:To study hongzhenzhu xuezhi kang capsule(HXKC) in pharmacodynamics for the sustainable utilization and development of the natural plant resources in the Qinghai-Xizang Plateau,such as Nitraria tangutorun Bobr.. OBJECTIVE:To probe into the mechanism of HXKC on regulating blood lipid,developed mainly by the natural plant fruits enriched in Winghai-Xizang Plateau and Chaidamu region,I.e.Nitraria tangutorun Bobr.,Hippophae rhamnoides L,Lycium barbarum L,etc.. DESIGN:Randomized controlled trial. SETTING,MATERIALS and INTERVENTIONS:The experiment was performed in the Pharmacological Experiment Room of Institute of Plateau Medical Science in Qinghai.66 Kunming mice and 68 Wistar rats of either sex were employed.According to the random number table,the mice were divided into normal forage breeding group of 12(for blank control) and high-lipid forage breeding group of 54.2 weeks later after breeding,the mice were divided at random into high-lipid model group,high dosage group(HXKC,2.0 g/kg),low dosage group(HXKC,1.0 g/kg) and positive control group(zhibituopian,1.05 g/kg).The medicine was given by gastric infusion 10 mL/kg and the same volume pipe water was provided in blank control and high-lipid model groups to determine serum total cholesterol(TC) and triacylglyceol(TG) content in mice.According to the random number table,68 normal rats were divided into blank control group(normal forage),high-lipid model group,HXKC low dosage group(0.5 g/kg),moderate dosage group(1.0 g/kg),high dosage group(2.0 g/kg) and positive control group(zhibituopian,1.05 g/kg).The medicine was given by gastric infusion 10 mL/kg and the same volume pipe water was provided in blank control and high-lipid model groups to determine serum TC,TG,high density lipoprotein cholesterol(HDL-C),MDA content and activity of superoxide dismutase( SOD) . RESULTS:Compared with high-lipid model group,serum TC,TG content in the rats and mice with hyperlipemia in HXKC high and low dosage groups were remarkably lower(P< 0.05- 0.01).Compared with high-lipid model group(8.64± 2.07) μ mol/L],serum MDA content in the rats with hyperlipemia in HXC high and moderate dosage groups were remarkably lower(6.17± 1.81) ,(6.89± 1.54) μ mol/L](P< 0.05- 0.01).Compared with high-lipid model group,the activity of erythrocyte SOD in the rats with hyperlipemia in HXC high,moderate and low dosage groups were remarkably higher(P< 0.05). CONCLUSION:To suggest that HXKC capsule acts on preventing hyperlipemia and improving atherosis.
